Abstract:
Biofilm structure and functionality depend on extracellular polymeric substances (EPS), but analytical methods for EPS often lack specificity which limits progress of biofilm research. EPS were extracted from aerobic granular sludge and analyzed with frequently applied colorimetric methods. The colorimetric methods were evaluated based on their applicability for EPS analysis. EPS fractions of interest were proteins, sugars, uronic acids and phenolic compounds. The applied methods (Lowry method, bicinchoninic acid assay, phenol sulfuric acid method, carbazole sulfuric acid method) were investigated in terms of their sensitivity towards the selected standard compound. Interference of compounds present in EPS with the colorimetric methods was further evaluated. All methods showed to be highly depending on the choice of standard compound and susceptible towards interference by compounds present in EPS. This study shows that currently used colorimetric methods are not capable of accurately characterizing EPS. More advanced methods are needed to be able to draw conclusions about biofilm composition, structure and functionality.

Abstract:
Door de groeiende wereldbevolking in combinatie met klimaatverandering en welvaartstoename zal de druk op de voedselvoorziening sterk gaan toenemen. Naast een verschuiving naar plantaardige eiwitten, moet de wetenschap op zoek naar alternatieve bronnen voor eiwitten. Door micro-organismen geproduceerde eiwitten, zogenaamde single cell proteins, vormen een alternatief met een grote potentie. Dit onderzoek richt zich op de biosynthese van single cell protein (SCP) door lithotrofe waterstofoxiderende bacteriën die koolzuurgas en ammonium onder invloed van waterstof als energiedrager en zuurstof op efficiënte wijze omzetten in celeiwit. Dit power-to-protein is ontwikkeld door Avecom in Gent. In 2014/15 is een inventariserende studie in het kader van TKI Watertechnologie uitgevoerd om de levensvatbaarheid van de koppeling van het Power to Protein- concept aan de afvalwaterketen in te schatten en deze te toetsen aan een casus voor de ‘urbane zone van Amsterdam, fase 1’ (Oesterholt, 2015). Uit de studie bleek een enorme potentie waarbij jaarlijks 6.300 ton eiwit kan worden geproduceerd uitgaande van ammonium in de natte fractie van de slibvergisting op RWZI Amsterdam-West. Hiermee zou voor 36 % van de Amsterdamse bevolking in de (netto) eiwitbehoefte kunnen worden voorzien. De economische potentie hang sterk af van de kosten voor de productie van de grondstoffen ammonium en waterstof én de verkoopwaarde van eindproduct.

Abstract:
Schimmels zijn efficiënte eiwitproducenten en prima toepasbaar voor microbiële productieprocessen. Het Bredase bedrijf BioscienZ ontwikkelt zo’n productieproces voor eiwitrijke vleesvervangers en het kippeneiwit ovalbumine en gebruikt hiervoor schimmels die groeien op aardappels en suikerbieten. Het begon allemaal een paar jaar geleden met een bezoek van De Vegetarische Slager, vertelt Wim de Laat, directeur van BioscienZ in Breda. Na jarenlang bij grote bedrijven zoals Gist-Brocades, DSM en Nedalco microbiële processen te hebben ontwikkeld, was De Laat in 2010 voor zichzelf begonnen, eerst als adviseur en in 2014 met een laboratorium voor contractonderzoek. De Laat had zich vooral gespecialiseerd in het ontwikkelen van processen op basis van schimmels.

Abstract:
What innovations reduce Europe’s dependency on unstable and contested imports of coconut oil, palm kernel oils, fatty acids and castor oil? For substitutes, researchers of Wageningen University and Research, by the COSMOS-project, looked into the crops Camelina and Crambe. New plant breeding techniques, such as CRISPR, enhanced characteristics of these crops regarding both their flavor and the molecular structure of their oils. One can further valorise the side streams, such as press cake, by using them as feed for the Black Soldier Fly larvae. The Entomology-department thus, out of these new crops, realise extra valuable proteins and insect oils.
