Abstract The present invention relates to an approach aimed at the modification of codons in individual polynucleotide sequences encoding a heterologous protein of interest, without altering the amino acid sequence of the polypeptide to enhance the amount of functional expression in a host organism of interest. In its broadest aspect, this approach exploits redundancy in the genetic code by providing a universal set of codons which may be used at certain positions in the polynucleotide sequence in order to achieve improved heterologous protein production in a range of host cells. The present invention also relates to specific codons which may be used to increase protein expression in particular hosts. The present invention also relates to the optimization of the translation efficiency of messenger RNAs on the basis of their secondary structure characteristics, and the provided set of criteria may be used to increase protein expression in particular hosts.
