Assuring safety without animal testing concept (ASAT). Integration of human disease data with in vitro data to improve toxicology testing

According to the Assuring Safety Without Animal Testing (ASAT) principle, risk assessment may ultimately become possible without the use of animals (Fentem et al., (2004). Altern. Lab. Anim. 32, 617–623). The ASAT concept takes human disease mechanisms as starting point and tries to define if activation of these mechanisms by chemical exposure in in vitro models can be used for toxicological risk assessment. The goal of the present research was to study if integration of public data, from human diseases and in vitro toxicology, is possible at the data level. Two human diseases, associated with chemical exposure, were included: human hepatocellular carcinoma and allergic contact dermatitis (ACD). Data were retrieved from expert knowledge and different online sources (e.g. GEO, ArrayExpress) and a Knowledge Base for storage and modelling of the data was established. Using the Knowledge Base for ACD, it was possible to discern sensitizing from non-sensitizing compounds, as defined by enrichment of clinically defined disease gene sets in in vitro genomics datasets. In addition, the strongest sensitizers most profoundly activated these gene sets. During the presentation, the Knowledge Base will be shown. In addition, the ongoing incorporation of (reverse) kinetic modelling to judge the relevance of in vitro concentrations, in relation to realistic in vivo exposure scenarios, will be illustrated. Finally, the expansion towards the development of data models for other disease areas (cholestasis) will be discussed.

Lactic acid bacteria are claimed to have immunomodulating effects. Stimulation as well as suppression of T helper (Th)1 mediated immune responses, have been described for various strains. Experiments involving Lactobacillus casei Shirota (LcS) detected mainly enhancement of innate immune responses and promotion of Th1 mediated immune reactivity. To confirm and further investigate modulation of Th1 responses and development of autoimmune disease by LcS, the consequences of oral administration of LcS were assessed in several experiments. The effect of LcS varied between the different models. No modulation was found in the mitogen-induced cell proliferation and cytokine release assays in mesenteric lymph nodes of Wistar rats. LcS inhibited the Th1 mediated immune response in an adapted murine Local Lymph Node Assay (LLNA) in BALB/c mice, whereas experimental autoimmune encephalomyelitis (EAE) in Lewis rats was aggravated. These varying effects on Th1 responses indicate that beneficial as well as harmful effects on immune related disorders could occur after LcS consumption. Since microarray analysis is suggested to be more sensitive and predictive than functional tests, gene expression profiling was included as an alternative endpoint in the testing of immunomodulation. The detected gene expression profiles did not reflect the effects of LcS on the immune system. Microarray analysis may therefore have no more predictive value than immune function assays when investigating immunomodulation by probiotics. To gain further insight into effects of probiotics on immune function, experiments including cytokine assays and gene expression analysis combined with disease models could be useful

Vanuit Europese regelgeving geldt per1 november 2005 de verplichting om de aanwezigheid van allergie opwekkende stoffen (allergenen) op de etiketten van voedingsproducten te vermelden. Om dit beleid goed te handhaven moet bekend zijn hoeveel allergenen een mens binnen mag krijgen voordat er een allergische reactie optreedt. Een extra factor om rekening mee te houden in allergenen onderzoek, zijn nieuw ontwikkelde soorten voeding ('novel foods'). Er kan bij deze voeding sprake zijn van nieuwe of onbekende eiwitten. Nu is nog moeilijk te voorspellen of deze eiwitten allergie opwekken

It was of importance in the evaluation of feedingstufrs to know its quantitative composition. A qualitative botanical indicated purity but a quantitative test was needed for the degree of purity. Natural feedstuffs always contained a certain amount of foreign material, which could be considered either as natural or chance contamination. The manufacturer had to ensure that commercial feedstuffs had a high degree of purity. Whenever there was consideration in the trade of standards for purity, it was necessary to be able to check these standards.

Ezendam worked out a method of measuring impurities both quantitatively and qualitatively. It was tested on some feedstuffs and thus led the way for microscopic study of feedingstuffs.
