Data-Driven Modeling of Intracellular Auxin Fluxes Indicates a Dominant Role of the ER in Controlling Nuclear Auxin Uptake

In plants, the phytohormone auxin acts as a master regulator of developmental processes and environmental responses. The best characterized process in the auxin regulatory network occurs at the subcellular scale, wherein auxin mediates signal transduction into transcriptional programs by triggering the degradation of Aux/IAA transcriptional repressor proteins in the nucleus. However, whether and how auxin movement between the nucleus and the surrounding compartments is regulated remain elusive. Using a fluorescent auxin analog, we show that its diffusion into the nucleus is restricted. By combining mathematical modeling with time course assays on auxin-mediated nuclear signaling and quantitative phenotyping in single plant cell systems, we show that ER-to-nucleus auxin flux represents a major subcellular pathway to directly control nuclear auxin levels. Our findings propose that the homeostatically regulated auxin pool in the ER and ER-to-nucleus auxin fluxes underpin auxin-mediated downstream responses in plant cells. Middleton et al. study how the plant phytohormone auxin enters the nucleus by using quantitative phenotyping in single plant cell systems and bespoke mathematical models that relate controlled perturbations to experimentally measurable responses. Their findings show that auxin predominantly enters the nucleus via the endoplasmic reticulum.

Deposits of peat underneath peat swamp forests are among the world’s largest reservoirs of carbon. Although tropical peatlands occupy only about 0.3 % of the global land surface, they could contain as much as 20 % of the global soil carbon stock, representing 63–148 Gt of carbon.

Peat swamp forests are among the worlds most threatened and least known ecosystems. In Southeast Asia large areas of peat swamp forest have been deforested, converted for agricultural projects or into plantations (such as oil palm).

Drainage through canalisation has frequently severely disrupted water table level dynamics, resulting in CO2 emissions due to oxidisation and vulnerability to fire, especially during ‘El-Niño’ years. Water management is essential in addressing disturbances and rehabilitation of degraded tropical peatlands.

Radar satellite observations can be made frequently, also in the wet season.Because of a certain level of penetration of the radar waves, also observation below the canopy is possible. Particularly the L-band sensors on board the former JERS-1 and ALOS-1 satellites are superior to all other spaceborne sensors for assessment of flooding and drought conditions and, thus, hydrological cycles.

New Arenas of Engagement at the Water Governance-Climate Finance Nexus? An Analysis of the Boom and Bust of Hydropower CDM Projects in Vietnam

This article explores whether new arenas of engagement for water governance have been created and utilised following the implementation of the Clean Development Mechanism (CDM) in large hydropower projects in Vietnam. Initial optimism for climate finance – in particular amongst Northern aid providers and private CDM consultants – resulted in a boom in registration of CDM hydropower projects in Vietnam. These plans, however, have since then busted. The article utilises a multi-scale and multi-place network governance analysis of the water governance-climate finance nexus, based on interviews with government officials, consultants, developers, NGOs, multilateral and international banks, and project-affected people at the Song Bung 2 and Song Bung 4 hydropower projects in Central Vietnam. Particular attention is paid to how the place-based nature of organisations shapes the ability of these actors to participate in decision-making. The article concludes that the CDM has had little impact on water governance in Vietnam at the project level in terms of carbon reduction (additionality) or attaining sustainable development objectives. Furthermore, whilst climate finance has the potential to open new, more transparent and more accountable arenas of water governance, current arenas of the water governance-climate finance nexus are 'rendered technical', and therefore often underutilised and inaccessible to civil society and project-affected people.
A continuum approximation to an off-lattice individual-cell based model of cell migration and adhesion

Cell–cell adhesion plays a key role in the collective migration of cells and in determining correlations in the relative cell positions and velocities. Recently, it was demonstrated that off-lattice individual cell based models (IBMs) can accurately capture the correlations observed experimentally in a migrating cell population. However, IBMs are often computationally expensive and difficult to analyse mathematically. Traditional continuum-based models, in contrast, are amenable to mathematical analysis and are computationally less demanding, but typically correspond to a mean-field approximation of cell migration and so ignore cell–cell correlations. In this work, we address this problem by using an off-lattice IBM to derive a continuum approximation which does take into account correlations. We furthermore show that a mean-field approximation of the off-lattice IBM leads to a single partial integro-differential equation of the same form as proposed by Sherratt and co-workers to model cell adhesion. The latter is found to be only effective at approximating the ensemble averaged cell number density when mechanical interactions between cells are weak. In contrast, the predictions of our novel continuum model for the time-evolution of the ensemble cell number density distribution and of the density–density correlation function are in close agreement with those obtained from the IBM for a wide range of mechanical interaction strengths. In particular, we observe ‘front-like’ propagation of cells in simulations using both our IBM and our continuum model, but not in the continuum model simulations obtained using the mean-field approximation.

Soil heat flux is difficult to measure accurately and soil heat flux plates are difficult to calibrate. In this research the reference heat flux was calculated from the temperature gradient and independent thermal conductivity measurements. Reference conductivities, as measured by the non-steady state probe method, have an error of about 2%, while the temperature gradient was measured to an accuracy better than 1%. This results in very reliable reference measurements. Compared with this reference, commercially available heat flux plates have significant inaccuracies. The 1 mm thin TNO PU 43 T sensor was the most accurate with an average relative error of only 4%. A promising new technique is the in situ self calibration, as performed by the Huksefluks HFP-01-SC disc sensor. With self calibration this sensor achieves an accuracy of 5% and confers several advantages for field use. The MIDDLETON CN3 and TNO WS 31S sensors had relative errors of about 20%. The ring shaped sensor Huksefluks SH1 gave relatively poor results, because it measured, in fact, the temperature gradient instead of the heat flux. The results of this sensor remained poor after correcting for the thermal conductivity of the sand. For all sensors the same conclusions hold for non-steady state conditions with evaporation. The often used Philip (1961) correction factor is shown to be not very accurate: in only half of our experiments its use decreased the relative measuring error; and in some cases it made it worse. However, the correction remains useful as a tool for designing soil heat flux sensors; also a positive relation exists between the magnitude of the correction and the inaccuracy of the measurement.
