The application of quantitative thin-layer chromatography (TLC) involving in situ spectrodensitometric measurements with a flying-spot densitometer is described in this study for the analysis of aflatoxin B 1 , quinine hydrochloride and L(+)-ascorbic acid (vitamin C) in various food and foodstuffs.

Several aspects dealing with the quantitative analysis of aflatoxin B 1 in peanuts and peanut products were investigated: isolation, thin-layer chromatographic conditions, visual estimations and fluorodensitometric: measurements. The results of this study contributed to the development of the Official method of analysis for aflatoxin B 1 in the Netherlands. A new spotting procedure for aflatoxin assay was introduced ("anti-diagonal" technique) improving the accuracy for visual B 1 estiminations for samples with low contamination levels (0-10 μg/kg).

An in situ fluorodensitometric procedure was developed for the quantitative determination of quinine hydrochloride in soft drinks containing quinine whereas in situ densitometric transmittance measurements were applied for the quantitative determination of L(+)-ascorbic acid (vitamin C) in buttermilk.

It was generally concluded that in situ spectrodensitometric procedures were suitable for quantitative routine determinations in food control work, for specific purposes.
