    Ieder jaar vallen er doden en gewonden in het verkeer bij ongevallen waarbij alcohol in het spel is. Het alcoholslotprogramma draagt bij aan het omlaag brengen van het aantal verkeersslachtoffers en is bedoeld voor bestuurders die met te veel alcohol op aan het verkeer hebben deelgenomen. Als onderdeel van het programma wordt een alcoholslot in het voertuig van deelnemers ingebouwd zodat zij leren omgaan met alcohol en verkeer. Bij toepassing van het alcoholslot kan het voertuig niet gestart worden voordat een geldige ademtest is afgelegd, ook kan onderweg door het apparaat om hertesten gevraagd worden. Omdat het alcoholslot mogelijk niet alleen het gebruik van ethanol detecteert maar ook melding zou kunnen geven na gebruik van andere stoffen is onderzoek gedaan naar het ontstaan van vals-positieve meetresultaten van het alcoholslot.

    Our environment is constantly threatened by large amounts and variations of man-made chemicals and natural substances. Parts of these substances accumulate and contaminate soil and surface water, affecting the organisms living in it and eventually contaminate the food chain. The European Union (EU) has imposed regulations and obliged EU member states to monitor for possible contaminants in the environment and food. For this, highly sophisticated mass spectrometry (MS) techniques, which can nowadays screen >100 contaminants in a single run, are applied. For rapid and inexpensive screening of contaminants, bioactivity-based screening assays are applied, however, identification of compounds based on their chemical-physical properties is not possible. As both methods cannot identify emerging and unknown bioactive contaminants, there is a need for new tools and concepts. In this thesis, new bioaffinity MS (BioMS) concepts, using an antibody, transport proteins and a receptor, are presented for the screening and identification of contaminants. In the first concept, monoclonal antibodies (Mabs) against ochratoxins were coupled to fluorescent labeled paramagnetic microbeads for high-throughput flow cytometric screening of ochratoxins in wheat and cereal. The identification of ochratoxins with nano-ultra performance liquid chromatography-quadrupole-time-of-flight-MS (nano-UPLC-Q-ToF-MS) was achieved in full scan accurate mass mode. In the second BioMS approach, the flow cytometer was replaced by UPLC-triple quadrupole (QqQ)-MS for rapid screening of thyroid transporter ligands. For this, thyroid transport protein transthyretin (TTR) was immobilized onto inexpensive non-colored paramagnetic microbeads and a stable isotopic thyroid hormone was used as label in the competitive inhibition format. For the identification of TTR-binding endocrine disrupting chemicals (EDCs) in process water and urine, nano-UPLC-Q-ToF-MS was used. In order to perform high-throughput screening, a microtiter plate-based high-throughput BioMS approach was developed with the same beads but coupled with recombinant human sex hormone-binding globulin (rhSHBG) for the detection of designer steroids in dietary supplements. Following the screening with rhSHBG-based BioMS using LC-QqQ-MS, the rhSHBG bioaffinity extracts were injected onto chip-UPLC-Q-ToF-MS operated in full scan mode and a wide range of steroids were identified. The same approach was applied with the estrogen receptor α (ERα) in which LC-QqQ-MS, instead of the commonly applied GC-MS, was used for the screening of estrogens with a suitable LC-MS-compatible label. The identification of estrogens in ERα-purified supplement extracts was achieved with UPLC-ion mobility (IM)-Q-ToF-MS. These new BioMS concepts present new tools for the screening and identification of emerging yet unknown food and environmental contaminants to ensure consumer’s health and fair play in sports.

    The aim of the present project was to determine the most critical steps in the production of fats and oils. First, production processes of vegetables oils, animal fat, fish oil, biodiesel and fat blending were studied and experts from the industry as well as in-house dioxin experts were consulted to identify the most critical steps. Then, monitoring data for vegetable oil, animal fat and fish oil were analysed to determine which products have an increased risk of dioxin contamination.

    New methods for the screening of antioxidants in three Sideritis species

    This thesis describes the rapid and robust evaluation of antioxidant activity of complex mixtures such as plant extracts. The study was directed to establish the best screening and isolation procedures for components with antioxidative properties to ensure the development of an “algorithm” for studying natural antioxidants. Three different screening methods (the b-carotene bleaching test (BCBT), the headspace GC method (HS-GC) and the off-line radical scavenging assay with the DPPH radical (DPPH method) and various extraction schemes were used. Three plants from the genus Sideritis grown in Bulgaria (S. scardica, S. syriaca and S. montana) served as test species. The strengths and limitations of each method were illustrated by testing a number of extracts of different polarity. A novel HPLC-DPPH method to rapidly and sensitively pinpoint individual antioxidants in complex mixtures with as little as possible fractionation procedures was developed. The use of the alternative ABTS·+ radical cation made the method in most cases even more sensitive. The instrumental set-up and physico-chemical parameters of both methods were studied. The methods were successful for qualitative and semi-quantitative measurements of pure antioxidants and extracts. A review is under preparation on the use of on-line methods. As a result, a range of flavonoid-, phenylpropanoid- and iridoid glycosides were isolated. The radical scavenging activity of the isolated compounds by off- and on-line DPPH methods was studied. Three novel compounds were isolated: flavonoid glycosides luteolin-4'-methylether-7-O-[6'''-O-acetyl-β-D-allopyranosyl-(1-2)-β-D-glucopyranoside and hypolaetin-4'-methylether-7-O-[β-D-allopyranosyl-(1-2)-6''-O-acetyl-β-D-glucopyranoside, and iridoid glycoside 3''-O-p-coumaroyl-6''-O-acetyl melittoside from Sideritis species. Preliminary studies by the HS-GC method were applied to investigate how Sideritis extracts and pure components would behave in real systems: bulk oils and oil-in-water emulsions. Conjugated dienes and hexanal formation were monitored using rosmarinic acid and BHT as standards. They showed good inhibitory effects with regard to dienes and hexanal formation in both test systems.

    The brominated flame retardants, hexabromocyclododecane (HBCD) and tetrabromobisphenyl-A (TBBP-A) were analysed in samples of human blood from 40 different individuals. In only one individual was HBCD detected. In about half of the individuals, TBBP-A was detected. When they were detected, the amounts of both chemicals were near the limits of quantification. That the chemicals were detected confirms that certain individuals were exposed to sources of these substances. The toxicological implications of these low levels are not well known. Very few studies of the toxicology of these compounds have been performed to date.
