    Proficiency testing is conducted to provide laboratories with a powerful tool to evaluate and demonstrate the reliability of the data that are produced. Next to validation and accreditation, proficiency testing is an important requirement of the EU Additional Measures Directive 93/99/EEC [1] and is required by ISO 17025:2005 [2]. The aim of this proficiency test was to give laboratories the possibility to evaluate or demonstrate their competence for the analysis of aflatoxins in pig feed.

    The added value of proficiency tests: choosing the proper method

    To gain insight into the different extraction processes used in laboratories in various European countries and the effect these methods might have on the number of Meloidogyne second-stage juveniles (J2) extracted from soil samples, a ring test with 2 elements was conducted. The first element used the Baermann funnel as a method performance test. The second element was a proficiency test, in which the laboratory’s standard technique was compared with the Baermann method. Chilled samples of 100 cm3 (10 per extraction method) were sent to participants in insulated containers. In the nematode counts distinction was made between Meloidogyne J2, other plant-parasitic nematodes and/or saprophytes. A total number of 18 participants took part in the method performance test and 13 institutes took part in the proficiency test using 11 different extraction techniques. The initial population density in the soil was 2025 J2 (100 cm3 soil)-1, determined by Oostenbrink elutriator and 4 weeks incubation. In the method performance test the institutes found Meloidogyne J2 numbers varied between 0 and 705 J2 (100 cm3)-1. Using the Baermann extraction method the median number per institute ranged from 0 to 377 J2 (100 cm3)-1. In the proficiency test comparison amongst the different methods showed an increase of more than 100-fold, with the highest median of 3733 J2 (100 cm3)-1 and the lowest of 34 J2 (100 cm3)-1; the difference was caused mainly by the incubation effect. For the other plant-parasitic nematodes and the saprophytes, the differences between the most and least efficient extraction methods were much smaller. The variance of the Meloidogyne counts was the highest for the standard Baermann extraction technique; other similar techniques also had high variances. The automated zonal centrifuge had the lowest variance. The Baermann method is not advisable for survey purposes

    An increasing number of reports confirm the world-wide presence of the perfluoroalkyl substances (PFASs). As a consequence, the demand for qualitative and quantitative environmental occurrence data requires accurate risk assessments. To improve the analytical quality of the determination of PFASs in food and environmental samples, a 4th international interlaboratory study (ILS) was conducted in 2011. A total of 31 partners participated, and, depending on the sample matrix, up to 29 data sets were submitted. The ILS focused on food samples, as it was organized by the PERFOOD consortium in collaboration with QUASIMEME. The results showed that the cumulative experience of the participants has improved their analytical quality over four international ILSs. Several sources of errors were identified and methods to avoid them are suggested.

    The design oriented research described in this thesis aims at designing an realizing an electronic performance support system for food chemistry laboratory classes (labEPSS). Four design goals related to food chemistry laboratory classes were identified. Firstly, labEPSS should avoid extraneous cognitive load caused by the instructional format of the laboratory classes. Secondly, labEPSS should let students prepare for their laboratory experiments. Thirdly, labEPSS should support the communication in the laboratory class between students and between students and supervisors. Fourthly, labEPSS should give students the freedom to plan their experiments, without supervisors losing control and without risking overbooking of equipment. To address these goals, a couple of tools were designed, realized and subsequently used and evaluated in two model food chemistry laboratory classes:

    A web-based laboratory manual, aiming to provide students with just-in-time procedural information (e.g. how an apparatus looks like, where chemicals can be found). A web-based experiment design tool, aiming to let students design their research strategy as a workflow beforehand and support students while carrying out this strategy in the laboratory. A ‘web-app’ for students’ smartphones providing the same functionalities as the digital laboratory manual. A web-based equipment booking system, which is part of the web-based experiment design tool.

    Based on the evaluations it can be concluded that students and supervisors appreciated the tools and that these tools are capable of reaching the design goals. Finally, an overall design of labEPSS is proposed, in which the tools offer an integrated experience. Because labEPSS is highly configurable, it can be used in many different laboratory classes throughout curricula.

    Are antibiotic screening approaches sufficiently adequate? A proficiency test

    A proficiency test including the screening analysis of antibiotics in beef using cryogenicly minced materials was organized by RIKILT in 2009. The test included blank beef samples and beef samples spiked with either flumequine or a combination of lincomycin and spectinomycin around the maximum residue limits [1]. The suitability of the materials was demonstrated with a homogeneity and a stability study. This study showed that cryogenically minced spiked muscle material is suited for proficiency tests aiming at the screening and the confirmatory analysis. Of the 26 participants, 23 carried out their in-house screening approach involving microbial, biochemical or instrumental methods, or a combination of these to cover the broad range of antibiotic groups. The false negative rate was 73% for microbial methods, 50% for biochemical and 22% for instrumental methods. These results indicate that substantial effort is needed to improve screening approaches and that more regular proficiency tests are needed to reveal the shortcomings in the currently applied screening methods.

    In november 2008 is door het bedrijfsleven in een tweetal partijen diervoedergrondstoffen (sojaschilfers/-meel) melamine aangetroffen op niveaus tussen 40 en 800 mg/kg. Naar aanleiding hiervan heeft de VWA aan RIKILT verzocht om deze monsters (genomen door het bedrijfsleven) te analyseren op melamine en daarnaast, ten behoeve van de risicobeoordeling, op de hydrolyseproducten ammeline, ammelide en cyanuurzuur

    In opdracht van RWS-RIZA is het project “Het concentreren en opwerken van waterige monsters volgens de XAD procedure” uitgevoerd. De XAD procedure is uitgevoerd volgens een protocol dat is gebaseerd op methoden ontwikkeld bij het RIVM, Laboratorium voor Ecotoxicologie te Bilthoven. Daarna zijn de waterige monsters naar TNO IMARES in Den Helder gegaan voor beoordeling van de toxiciteit.

    In de Kaderwet Diervoeders van 2003 is o.a. beschreven hoe toezicht en controles op diervoeders uitgevoerd moeten worden. Een aspect ervan is hoe bemonstering en analyse van de partij diervoeder dient plaats te vinden. De regelgeving in de Kaderwet Diervoeders en Besluit Diervoeders is op een aantal punten vrij algemeen. Er worden meestal door de ambtenaar drie eindmonsters gemaakt: een monster voor analyse, een contramonster en een bedrijfsmonster. In de praktijk blijkt echter dat het niet altijd duidelijk is welk van deze monsters precies het contramonster is, waar het naar toegestuurd en waar het geanalyseerd kan worden. Om hierop een duidelijk antwoord te krijgen zijn de van toepassing zijnde wetgeving en overige relevante documenten bestudeerd. Met name de Verordening EG/882/2004 geeft veel helderheid in de geschetste problematiek.

    The European eel ( Anguillaanguilla L.) is a catadromic fish species with its spawning grounds thousands of kilometers away in the ocean, possibly theSargasso Sea. The objective of this study was to elucidate this oceanic phase of migration for the European eel (Anguillaanguilla L.) in the laboratory.

    This thesis focuses on several aspects of the life cycle of the European eel:

    A)   Preparation to migration, B)    Simulated migration,

    C)   Effect of environmental factors (viruses and PCB's) on the migration,

    D)   Effect of swimming on maturation.

    In order to investigate this we simulated the migration of spawners in the laboratory by building 22 large swim tunnels of 127 liter specially developed for long term migration. Therefore the title of this thesis is: 'Simulated migration of European eel' . What matters is how much energy is required for the crossing of the Atlantic Ocean. An eel uses only 58 g fat/kg fish while a salmon uses 300 g fat/kg. The energycost of transportation (COT) are4-6 times lower for eels than for salmonids. The second aims of the investigations were to study environmental factors that may have impact on the migration capacity like viruses and pollutants (PCB's). The third aims of the investigations were to test whether endurance swimming induces sexual maturation and development of the gonads.

    We can conclude that European eels are very efficient swimmers and that healthy well fed eels are able to reach theSargasso Sealeaving enough reserves for the reproduction. Endurance swimming of eels and the ability to migrate are negatively influenced by infections with viruses like EVEX. Also PCB's which are released from depleted lipid stores after migration may interfere with energy use and metabolism.

    So, although we have found several habitat factors that may interfere with fitness and endurance swimming such as viruses and toxicants (PCBs) which could be a contributing factor to declining eel populations we found two positive results which in future studies may be of importance for reproduction of eels under artificial conditions. a) First, we found strong evidence that swimming triggers silvering and early maturation, b) Second, we observed for the first time in hormone treated eels that group spawning was collective and simultaneous.

    Volgens de richtlijnen van de EU Directive wordt de aanwezigheid van M. chitwoodi en M.fallax in partijen voortkwekingsmateriaal vastgesteld aan de hand van de zichtbare symptomen. Er is behoefte aan betere methoden om de aanwezigheid en de intensiteit van beide aaltjes in percelen en partijen vast te stellen. Dergelijke methoden, zoals eerder ontwikkeld voor de quarantaine-nematoden G. rostochiensis en G. pallida, kunnen dienen als basis voor de uitvoering van fytosanitaire taken van de PD en als grondslag voor Beslissing Ondersteunende Systemen voor de beheersing van nematoden op percelen.     Voor de periode 1 december 2002 tot 1 december 2003 is een Minas-balans opgesteld voor de nertsenfarm op 'Het Spelderholt' in Beekbergen. Er is zowel een verfijnde als een forfaitaire mineralenbalans opgesteld, op basis van een groot aantal monsters van de drijfmest uit vier mestkelders en de strooiselmest uit drie sheds. Aan de hand van 14 mestanalyses bleek het mogelijk om uit te komen op een zo goed als sluitende balans (fosfaatoverschot 2 kg; stikstofoverschot 36 kg). Daarnaast is een kleine ringtest gedaan naar de analyseresultaten van enkele Minas-laboratoria. Er blijken grote verschillen te zijn. Blijkbaar is het analyseren van nertsendrijfmest erg lastig, wat kan leiden tot onterechte heffingen

    Biobanks correspond to different situations: research and technological development, medical diagnosis or therapeutic activities. Their status is not clearly defined. We aimed to investigate human biobanking in Europe, particularly in relation to organisational, economic and ethical issues in various national contexts. Data from a survey in six EU countries (France, Germany, the Netherlands, Portugal, Spain and the UK) were collected as part of a European Research Project examining human and non-human biobanking (EUROGENBANK, coordinated by Professor JC Galloux). A total of 147 institutions concerned with biobanking of human samples and data were investigated by questionnaires and interviews. Most institutions surveyed belong to the public or private non-profit-making sectors, which have a key role in biobanking. This activity is increasing in all countries because few samples are discarded and genetic research is proliferating. Collections vary in size, many being small and only a few very large. Their purpose is often research, or research and healthcare, mostly in the context of disease studies. A specific budget is very rarely allocated to biobanking and costs are not often evaluated. Samples are usually provided free of charge and gifts and exchanges are the common rule. Good practice guidelines are generally followed and quality controls are performed but quality procedures are not always clearly explained. Associated data are usually computerised (identified or identifiable samples). Biobankers generally favour centralisation of data rather than of samples. Legal and ethical harmonisation within Europe is considered likely to facilitate international collaboration. We propose a series of recommendations and suggestions arising from the EUROGENBANK project.
