    This thesis focuses on a modern analytical method, proteomics, to investigate its use in the field of toxicological research. Proteomics is a high resolution method which separates all proteins present in a sample at a clearly defined state and compares this pattern to another one, under slightly different conditions (e.g. after exposure to a chemical). Protein changes may give rise to or reflect disease/harm of the individual and can be attributed to alterations in body functions/regulation systems. Analysis conditions and different varieties of proteomic methods are explained, and a brief introduction given where proteomics is already applied in toxicology. A specific investigation has been performed with the flame retardant HBCD (i.e. hexabromocyclododecane). It is a compound that accumulates in lipid tissue from where it is only slowly removed. Its mechanism of action is not yet completely understood and sometimes seems to be contradictory. Rats were exposed to HBCD in very low doses for just one week and liver proteins were compared to those of unexposed animals. As HBCD is suggested to disturb the thyroid system, both healthy and hypothyroid rats were investigated, of both genders. In female rats, not in males, some specific liver protein changes were seen in glucose/carbohydrate and lipid metabolism, and also in some stress related proteins. Changes were not dependent on the thyroid function of the females. These results are in line with previous findings that female rats were more susceptible to HBCD than males. In a further step, protein patterns of unexposed animals of both genders were compared, revealing gender-dependent differences that exceeded the effects seen in any of the other comparisons, mainly in the pathways that were also affected by HBCD in females. A previous proteomic study on serum proteins has also shown clear gender-dependent concentration differences in rats. This underlines the importance of performing studies both in female and male individuals. The detection of considerable gender-dependent protein alterations confirms that proteomics is a biochemical tool with high sensitivity and large potential also in toxicological research.

    Food security is one of the main issues in Ethiopia. Large amounts of money are spent on food security programs set up by federal- and regional governments and other organisations. Decisionmakers, as well as researchers, share a dependency upon reliable data on soils, water and crops. The authors of this report believe that the data from the Ethiopian soil laboratories are not reliable enough for decision-making purposes. Ethiopian laboratories need to improve their analytical services. The main problems concern the organisational set-up, and not merely a lack of budgets for facilities. Apart from organisational problems, managerial-, human resources-, technical-, instrumental- and supply issues were identified.

    Proficiency testing is conducted to provide laboratories with a powerful tool to evaluate and demonstrate the reliability of the data that are produced. Next to validation and accreditation, proficiency testing is an important requirement of the EU Additional Measures Directive 93/99/EEC [1] and is required by ISO 17025:200. The proficiency test for tropane alkaloids was organized by RIKILT, Wageningen UR in accordance with ISO 17043. The primary goal of this study was to provide laboratories with the opportunity to implement a method in their laboratory and to evaluate or demonstrate their performance regarding quantitative analysis of tropane alkaloids in cereals intended for human consumption and in animal feed.

    This report describes the activities employed in 2012 by RIKILT as National Reference Laboratory (NRL) in the field of dioxins and polychlorinated biphenyls (PCBs), pesticides in animal products, mycotoxins, heavy metals, polycyclic aromatic hydrocarbons (PAHs), and marine toxins according to Regulation (EC) No 882/2004 and Council Directive 96/23/EC. For each domain, the EURL-NRL network is described and interactions and meetings within the network are reported. In 2012 RIKILT attended EURL meetings for each domain. In the context of independent quality assurance, method validation and method comparison, RIKILT participated in profienciency tests and in interlaboratory validation and comparison studies. The results are reported and discussed. This report also summarises some of the scientific research supporting the NRL activities as well as other analytical activities within other projects that contribute to maintaining the expertise in the respective domains.

    Om met zekerheid vast te kunnen stellen dat er geen negatieve effecten optreden op het mariene milieu n.a.v. de oever-verdediging in Ooster- en Westerschelde is door Rijkswaterstaat in 2009 een monitoringsprogramma opgezet. Vanwege kostenoverwegingen zijn monitoringsactiviteiten in 2012 ingeperkt. Wel zijn op de Zeelandbrug (Oosterschelde) mosselen (Mytilus edulis) en Japanse oesters (Crassostrea gigas) bemonsterd om inzicht te verkrijgen in gehalten aan zware metalen tweeëneenhalf tot drie jaar na het aanbrengen van de vooroeververdediging. Dit rapport beschrijft het resultaat van de verschillende toxiciteitstesten die uitgevoerd zijn in het laboratorium van IMARES Den Helder. Daarnaast worden de resultaten van de metaalanalyses in mosselen en oesters bemonsterd op de Zeelandbrug besproken waarbij ingegaan wordt op de jaarlijkse variatie in metaalconcentratie een mogelijke trend in de tijd.

    The Europe Media Monitor (EMM) is a news gathering engine which is operated by the Joint Research Centre (JRC). EMM analyses news items published on the WWW, extracts relevant information, aggregates the collected information, issues alerts, and produces visual presentations of the information collected. Within the framework of the present study, we have analysed the potential of EMM to find and collect information on the WWW on GMOs that have not yet been assessed for their food/feed and environmental safety. Furthermore, we have analysed the current EMM GMO filters to see whether it is possible to improve their performance.

    In november 2008 is door het bedrijfsleven in een tweetal partijen diervoedergrondstoffen (sojaschilfers/-meel) melamine aangetroffen op niveaus tussen 40 en 800 mg/kg.     New preparation of fish material for interlaboratory study on PFCs

    The Institute for Environmental Studies, Vrije Universiteit (IVM) has requested Wageningen IMARES to prepare a new fish material for use in the interlaboratory performance study on analysis of perfluorinated compounds (PFCs) due to the low amount of contaminants in the previously prepared material. The new material was requested to be prepared from fillet of flounder and spiked with artificial mixture prepared from material provided by IVM. Homogenized material should be then packed in glass jars, sterilized and delivered to IVM. This document provides report on the preparation of material.

    Voor de teelt van Zantedeschia wordt veel gebruik gemaakt van plantmateriaal dat is vermeerderd via weefselkweek. Sinds de introductie van planten vermeerderd via weefselkweek zijn in toenemende mate afwijkingen in plantmateriaal waargenomen die worden toegeschreven aan de weefselkweekvermeerdering. Het gaat daarbij om afwijkingen zoals bossigheid, bont en extreem smal blad en bladverruwing. In dit onderzoek is het effect van een aantal behandelingen in weefselkweek op afwijkingen onderzocht.

    In The Netherlands a large quantity of domestic, agricultural and industrial organic wastes is produced. After purification or composting processes these substances can be partly re-used as organic fertilisers on the condition that this does not lead to contamination of the environment. In the so called Dutch regulation "Besluit kwaliteit en gebruik Overige Organische Meststoffen" (BOOM) (1) sewage sludge, compost and soil are considered as organic fertilisers. For these fertilisers the regulation sets maximum residue limits for the heavy metals cadmium, chromium, copper, mercury, lead, nickel, zinc and arsenic. By BOOM, normalised methods (NEN) are prescribed for the determination of these elements. RIKILT organised as supervisor of the regulation BOOM an interlaboratory study to check the performance of the laboratories involved in BOOM analysis

    Verslag van een onderzoek naar de mogelijkheden van indirect selectie in lelie en tulp met moleculaire merkers. De conclusie is dat het mogelijk is om in lelie en tulp om moleculaire merkers gebaseerd op de AFLP technologie te vinden en hiermee koppelingskaarten op te stellen. - Koppeling met belangrijke eigenschappen in lelie en tulp kan gevonden en toegepast worden. - Ombouw van AFLP tot ‘makkelijke’ PCR merker (SCAR of CAPS) is moeilijk, maar niet onmogelijk. Een aan te bevelen strategie voor de toekomst is om meerdere merkers hiervoor te kiezen en je niet te beperken tot één. Recent ontwikkelde, moderne technieken geven de mogelijkheid de benodigde sequentiegegevens sneller en nauwkeuriger te produceren. De geïdentificeerde Fusarium merkers in lelie lenen zich hier bij uitstek voor.
    Laboratory animal allergy : allergen exposure assessment and epidemiological study of risk factors

    The main objective of the study presented in this thesis was to estimate the prevalence rate of laboratory animal allergy and to determine its association with risk factors, like allergen exposure level, atopy, gender and other host factors. A cross-sectional survey was undertaken among 540 workers at 8 laboratory animal facilities. All participants completed a questionnaire and underwent skin prick testing with common and occupational allergens. Total and specific IgE measures were obtained. Prevalence rates of allergic symptoms (chest tightness (asthma), eye/nose and/or skin) due to working with rats or mice were 19% and 10%, respectively. The most common symptoms were eye/nose symptoms, 17% and 9% and asthmatic symptoms were found in 6% and 3% of the workers, respectively. The prevalence rate of sensitisation to rat or mouse allergens was 18% and I I%. Symptoms and sensitisation were strongly correlated. However, not all symptoms seemed to be IgE mediated. Rat and mouse allergy, defined as allergic symptoms accompanied by sensitisation, was highly associated with elevated total IgE (≥100 kU/1) and positive skin prick response to common allergens. The relationship between rat and mouse allergy and positive skin prick response to common allergens, could be completely explained by a specific response to cat or dog fur allergens.

    Stationary and personal air sampling was performed to identify determinants of exposure and estimate the animal allergen exposure. Animal caretakers experienced the highest exposure to aeroallergens. However, large variation within each job title was present. This may result from the wide range of tasks performed, of which handling contaminated bedding or conscious rats or mice showed the highest exposure. A relationship between allergen exposure and rat allergy became visible after excluding workers with four or more years of exposure to laboratory animals. This relationship was clearly visible for the atopic workers (having an pet allergy and/or elevated total serum IgE). The effect of exposure on rat allergy varied between atopic and non-atopic workers. Atopy should therefore be considered as an effect modifier of exposure. A higher prevalence rate of rat allergy was also found for men and smokers. However, these associations were not statistically significant.

    In this thesis, the development of genetically uniform strains of common carp, Cyprinus carpio L. is described. As in research on mammals, the use of genetically uniform fish could increase the quality (replicability, reproducability and repeatability) of experiments. Inbreeding was done by gynogenetic and androgenetic reproduction. With these methods, fully homozygous. offspring are produced in only one generation. The principle of these reproduction techniques is to eliminate the genetic contribution of one of the parents by (UV) irradiating the sexual products, followed by artificially inducing diploidy by physical shocks in the developing zygotes. Inbred strains, congenic strains and F1 hybrids were produced. Congenic strains were used to study sex determination in carp. Inbred strains and F1 hybrids were used to study the genetic control of gonad development after a quantitative genetic model had been developed to facilitate estimation of genetic variance. We show that genetically uniform carp strains are suitable as experimental animal model in genetic research and in bioassays (toxicological research). It is obvious that other research areas could also profit from genetically uniform carp strains.
