    The use of double (w1/o/w2) emulsions, in which part of the oil is replaced by small water droplets, is a promising strategy to reduce oil content in food products. For successful applications, (1) significant levels of fat reduction (i.e. significant amounts of water inside the oil droplets) have to be achieved, (2) double emulsions have to be stable against conditions encountered during processing and storage, and (3) the mouthfeel and sensory perception have to be similar to that of full-fat equivalents. With the present work, significant progress was made in understanding the complex relations between double emulsion design, achievable levels of fat reduction, emulsion stability and sensory perception. We show that through careful emulsion design, stable double emulsions with high levels of fat reduction (up to 50%) can be obtained while maintaining fat-related sensory properties, making double emulsions a promising approach for the development of fat-reduced food products.

    The fatter the better : selecting microalgae cells for outdoor lipid production

    In chapter 1 we introduce microalgae, photosynthetic microorganisms with potential to replace commodities (such as food, feed, chemicals and fuels). Production costs are still high, reason why microalgae are still only economically feasible for niche markets. We suggest to borrow the concept of plant domestication to select industrial microalgae cells. Two approaches can be successfully used to domesticate microalgae: adaptive laboratory evolution (ALE) and fluorescence assisted cell sorting (FACS). ALE takes advantage of the natural adaptability of microorganisms to different environments, while FACS actually select cells with specific phenotypes. This thesis aimed to select cells of Chlorococcum littorale with improved phenotypes, assuming that these cells could establish new populations with increased industrial performance.

    In Chapter 2 we wanted to know what happened during time to biomass and lipid productivities of Chlorococcum littorale repeatedly subjected to N-starvation. We tested 2 different cycles of N-starvation, short (6 days) and long (12 days). Short cycles didn’t affect lipid productivity, highlighting the potential of C. littorale to be produced in semi-continuous cultivation. Repeated cycles of N-starvation could have caused adaptations of the strain. Hence, we also discussed the implications of using repeated N-starvation for adaptive laboratory evolution (ALE) experiments. Chapter 3 introduces a method to detect and to select microalgae cells with increased lipid content. The method requires only the fluorescence dye Bodipy505/515 dissolved in ethanol, and the method was designed to maintain cellular viability so the cells could be used to produce new inoculum. In chapter 4 we evaluated a question that emerged while deciding which criteria to use to sort lipid-rich cells: does cellular size affects lipid productivity of C. littorale? We hypothesized that cells with different diameters have different division rates, which could affect lipid productivity. Therefore, we assessed the influence of cell diameter, as a sorting parameter, on both biomass and lipid productivity of Chlorococcum littorale (comparing populations before and after sorting, based on different diameters). Results showed that the size of vegetative cells doesn’t affect the lipid productivity of C. littorale. In chapter 5 we present a strategy to sort cells of C. littorale with increased TAG productivity using the method developed at chapter 3. Both the original and the sorted population with the highest lipid productivity (namely, S5) were compared under simulated Dutch summer conditions. The results confirmed our data from experiments done under continuous light: S5 showed a double TAG productivity. Our results showed also that the selected phenotype was stable (1.5 year after sorting) and with potential to be used under industrial conditions. In chapter 6 we extrapolated our results (indoor and outdoor) to other climate conditions. We ran simulations changing the light conditions to four different locations worldwide (the Netherlands, Norway, Brazil and Spain) to estimate both biomass and TAG productivities. Results indicated that biomass yields were reduced at locations with higher light intensities (Brazil/Spain) when compared with locations with lower light intensities (Norway/Netherlands). Hence, the choice of location should not be based on light intensity, but on how stable irradiation is. Chapter 7 is the general discussion of the thesis, demonstrating that both ALE and FACS are effective approaches to select industrial microalgae cells. We also present our view on how ALE and FACS could further improve microalgae strains for industry.

    Een goed voorbeeld van een bulkproduct dat gewonnen kan worden uit microalgen is triacylglycerol (TAG), een vet dat bestaat uit een glycerol molecuul met daaraan 3 vetzuurketens. Om de productie van TAG te kunnen verhogen is begrip van het biologische mechanisme dat TAG vorming veroorzaakt onontbeerlijk. Het doel van dit onderzoek was dan ook om meer inzicht te krijgen in het mechanisme van TAG accumulatie in microalgen die blootgesteld zijn aan nadelige groeicondities, en om de verkregen kennis te vertalen naar optimale kweekcondities voor TAG productie.

    Background and Aim

    The current overconsumption of sugar, salt and fat has been linked to the incidence of health problems. For this reason, it is necessary to decrease the intake of these food components. In the context of reducing the contents of sugar, salt and fat in food products while maintaining sensorial quality, the aim of this thesis was to identify and understand the factors that affect the perception of taste and fat-related sensory attributes in semi-solid foods. To achieve this aim, the relationship between the spatial distribution of sucrose, the mechanical properties of semi-solid gels and the perception of taste was investigated in detail. In addition, the relationship between the spatial distribution of fat and the perception of fat-related sensory attributes was investigated.

    Methods

    Model systems composed of layers of mixed agar-gelatin gels were used to control the spatial distribution of sucrose and fat. Layers varying in concentration of sucrose or oil-in-water emulsion droplets (i.e. fat) were combined to prepare gels with homogeneous and inhomogeneous distributions of sucrose and fat. A series of experiments were carried out using 2-Alternative Forced Choice (2-AFC) tests, line scale ratings and time-intensity analysis to investigate the effects of modulation of the spatial distribution of sucrose and modulation of the mechanical properties of gels on sweetness intensity. The relationship between the spatial distribution of tastants and consumer preference was investigated in different types of foods (e.g. semi-solid gels, breads and sausages). Furthermore, a Quantitative Descriptive Analysis (QDA) panel was carried out to investigate the effect of modulation of the spatial distribution of fat on the perception of fat-related sensory attributes.

    Results

    An inhomogeneous distribution of sucrose at large sucrose concentration differences enhanced sweetness intensity. The frequency of high-intensity discontinuous stimulation of taste receptors, which is related to the presence of tastant concentration differences in the mouth during oral processing, was identified as the driving factor for taste enhancement. The oral breakdown of semi-solid gels, which is affected mainly by fracture strain, and the mixing behavior of gel fragments formed upon breakdownwere shown to influence the frequency of receptor stimulation and, consequently, taste enhancement. Furthermore, the optimum frequency of high-intensity discontinuous stimulation required to maximize taste enhancement was shown to depend on the textural properties of the gel. These findings suggest that the spatial distribution of tastants and the mechanical properties (especially fracture strain) can be modulatedto maximize taste enhancement. Additionally, consumer preference was found to be maintained or increased in products containing tastants heterogeneously distributed in the food matrix.

    Fat-related mouthfeel attributes, such as spreadable and melting, were enhanced by an inhomogeneous distribution of fat at large fat concentration differences. The presence of high-fat zones on the surface of the model system was shown to maximize the enhancement of attributes related to fat. Therefore, the enhancement of fat-related attributes seems to be driven by the presence of high-fat zones preferably on the surface of the product rather than by the presence of fat concentration differences within the product.

    Conclusions

    The combined effects of modulation of the spatial distribution of tastants and fat and modulation of the mechanical properties can be applied in the development of low-sugar, low-salt and low-fat food products with improved taste quality.

    An experiment with 141 multiparous pregnant sows was conducted to investigate whether the supply of glucogenic energy at the end of pregnancy may be insufficient for an optimal growth of the foetuses in sows that are fed a diet with a high level of non-starch polysaccharides (NSP) and low level of starch. Sows were fed one of three diets from day 85 of pregnancy until parturition: : :Sows were fed a high NSP low starch diet. The daily intake of starch and sugar was 500 g. : :Sows were fed the same diet in the same amount as the sows in treatment 1 plus an extra daily amount of 500 g starch from wheat starch (is glucogenic energy). This means that the sows were fed 0.58 kg/d extra wheat starch (is 6.73 MJ glucogenic NE) during the last month of pregnancy. : :Sows were fed the same diet in the same amount as the sows in treatment 1 plus an extra daily amount of 200 g soya oil (is 6.73 MJ lipogenic NE). This treatment was added because otherwise it will not be clear whether energy or glucogenic energy is thelimiting factor. : :From day 1 of day 85 of pregnancy all sows were fed the high NSP low starch diet. During lactation all sows were fed the same lactation diet. Effects on litter performance, on changes in body weight and backfat thickness of the sows, and on feed intakeduring lactation were studied. The supply of extra dietary starch or extra dietary fat in late-pregnant sows that were fed a high NSP low starch diet did not increase litter weight at birth. This suggests that sows that were fed the high NSP low starch diet received sufficient glucogenic energy for foetal development. : :The number of stillborn piglets was numerically higher in sows that received extra dietary fat at the end of pregnancy. Sows that were fed extra dietary starch or extra dietary fat increased more in weight and backfat thickness during late pregnancy than sows that were fed the high NSP low starch diet. Decreases in weight and backfat thickness during lactation were similar for the three experimental treatments. Voluntary feed intake during lactation was slightly decreased in sows that were fed extra starch or extra fat in late pregnancy. :The present study shows that late-pregnant sows can be fed a high NSP low starch diet without adverse effects on litter weight at birth. A daily intake of 500 to 600 g starch and sugar in latepregnant sows seems sufficient for optimal growth of the foetuses.

    The stability of recombined milk fat globules

    The stability of the fat globules in recombined milk products against creaming, flocculation, clustering, partial coalescence and real coalescence, with the emphasis on partial coalescence, was studied. (partial) Coalescence was characterized by determining changes in globule size distribution and fat content. Without crystals the emulsions were mostly stable at rest and during flow. If crystals were present, natural cream and emulsions of milk fat-in-whey were unstable in a flow, while emulsions of milk fat and skimmilk or milk fat and buttermilk remained fairly stable and only gave partial coalescence if high shear rates were applied to emulsions with a high fat content and a large average diameter.

    In some cases partial coalescence resulted in the formation of a few large clumps that coalesced into floating fat upon heating the emulsion to above the melting point of milk fat thus causing a reduction of the fat content of the underlying emulsion, in other cases partial coalescence resulted in the formation of many small clumps that coalesced into larger fat globules upon heating, thus causing an increase in average globule size. Coalescence course and rate appeared to depend on emulsion type and applied treatment. Partial coalescence of milk fat-in-whey emulsions nearly always resulted in a decrease of the fat content. With a model starting from a small fraction of reactive globules gradually growing into clumps during the treatment, the coalescence process of these emulsions was fairly good quantitatively described. The ideas whereupon this model is based were used to qualitatively explain the different partial coalescence processes observed with the other emulsions.

    Doel van dit onderzoek is: Ontwikkeling van een methode voor het aantonen van gefraktioneerd botervet. Voor het aantonen van gefraktioneerd botervet zijn de analysen van de vetzuursamenstelling, vetzuren in 2 positie, cholesterolgehalte en triglyceridesamenstelling onvoldoende. Een meer betrouwbare detektiemethode werd ontwikkeld, gebaseerd op de kristallisatie van vnl. de verzadigde hogere triglyceriden in hexaan bij 12,5°C. Normaal botervet geeft daarbij geen kristallisatie terwijl gefraktioneerd botervet tot 15% kristalmassa oplevert. Indien meer dan 0,5% kristallen worden gevonden dan is de aanwezigheid van gefraktioneerd botervet duidelijk aangetoond. Met in principe dezelfde methode is aantonen van de zachte fraktie mogelijk.

    Verbetering van de analysemethodiek voor de bepaling van het gehalte aan steroltracers in EEG boter. Aan een botervet werd een bekende hoeveelheid stigma- respectievelijk sitosterol toegevoegd overeenkomstig de EEG-regeling. De zuiverheid van deze tracers werd gaschromatografisch vastgesteld via een interne standaard. Uit het monster botervet en de monsters botervet met toevoegingen werden volgens de huidige methode de sterolen geisoleerd door ze neer te slaan met digitonine (volgens NEN 6350). Om te onderzoeken of alle sterolen neergeslagen werden, werd uit het filtraat het onverzeepbare deel geextraheerd en onderzocht op sterolen (enzymatisch) en sterolsamenstelling. De sterolsamenstellingen werden vastgesteld, middels capillaire gaschromatografie. Daar vooral de stigmasterol tracer niet teruggevonden werd en bovendien een grote spreiding in de analyse gevonden werd, mede veroorzaakt door discriminatie van stigmasterol t.o.v. cholesterol bij het neerslaan met digitonine, werd besloten de sterolsamenstelling te bepalen via het onverzeepbare deel al dan niet na dunnelaagscheiding. Tenslotte is de spreiding van de gaschromatografische analyse onderzocht. Met massaspectrometrie werd de structuur bepaald van de component die met capillair GC naast B-sitosterol werd gevonden.

    Inventariserend onderzoek naar het voorkomen, zowel kwalitatief als kwantitatief, van polychloorbifenylen in vetbiopten. De vetbiopten zijn genomen bij studenten van de vakgroep voeding van de Landbouwhogeschool te Wageningen. In het kader van een samenwerkingsverband tussen het RIKILT en de LH te Wageningen zijn een aantal monsters menselijk weefsel onderzocht op het voorkomen van polychloorbifenylen. Een en ander is uitgevoerd teneinde inzicht te krijgen in de besmetting van menselijk vet met individuele polychloorbifenylen zowel qua identiteit als qua kwantiteit.
