Evoked Consumption Context Matters in Food-Related Consumer Affective Research

In this chapter we provide a brief overview of the research using scenarios to evoke context in affective (specifically emotion) research within the field of Sensory and Consumer Science. We then present two case studies that extend previously used scenarios and their application to other affective variables, such as consumption enjoyment. We demonstrate that consumers are capable of using scenarios to evoke complex situations, which involve not only environmental or temporal factors but also behaviors. The potential that these strategies have are then discussed and their advantages and limitations to other approaches such as virtual reality compared. We conclude that the underlying success factor of means of context evocation lies in the familiarity and appropriateness of the imagined situation, which becomes most relevant when collecting affective data in laboratory settings.

The complex challenges arising from climate change that exceeds the +2 °C target (termed ‘high-end climate change’) in Europe require new integrative responses to support transformations to a more sustainable future. We present a novel methodology that combines transition management and high-end climate and socioeconomic change scenarios to identify pathways and move Europe closer to sustainability. Eighteen pathways have been co-created with stakeholders through a participatory process. The pathways support Europe in moving towards a desirable future vision, through top-down and bottom-up actions that lower greenhouse gas emissions, reduce impacts of and vulnerabilities to climate and socioeconomic changes and enhance well-being. Analysis shows that the pathways that are robust to future scenario uncertainty are those that shift Europe towards sustainable lifestyles, support and strengthen good governance for sustainability and promote adaptive resource management for water, agriculture and energy. The methodology can support the design of the urgent actions needed to meet the requirements of the Paris Agreement and to transform Europe, in preparation for an uncertain future.

Stomatal cell lineage is an archetypal example of asymmetric cell division (ACD), which is necessary for plant survival1-4. In Arabidopsis thaliana, the GLYCOGEN SYNTHASE KINASE3 (GSK3)/SHAGGY-like kinase BRASSINOSTEROID INSENSITIVE 2 (BIN2) phosphorylates both the mitogen-activated protein kinase (MAPK) signalling module5,6 and its downstream target, the transcription factor SPEECHLESS (SPCH)7, to promote and restrict ACDs, respectively, in the same stomatal lineage cell. However, the mechanisms that balance these mutually exclusive activities remain unclear. Here we identify the plant-specific protein POLAR as a stomatal lineage scaffold for a subset of GSK3-like kinases that confines them to the cytosol and subsequently transiently polarizes them within the cell, together with BREAKING OF ASYMMETRY IN THE STOMATAL LINEAGE (BASL), before ACD. As a result, MAPK signalling is attenuated, enabling SPCH to drive ACD in the nucleus. Moreover, POLAR turnover requires phosphorylation on specific residues, mediated by GSK3. Our study reveals a mechanism by which the scaffolding protein POLAR ensures GSK3 substrate specificity, and could serve as a paradigm for understanding regulation of GSK3 in plants.

We compared the primary molt of the 4 species of skuas and jaegers (Stercorariidae) that breed in the Northern Hemisphere: Long-tailed Jaeger (Stercorarius longicaudus), Parasitic Jaeger (S. parasiticus), Pomarine Jaeger (S.
pomarinus), and Great Skua (S. skua). We analyzed primary molt data of 1,573 individuals of multiple age classes, mostly collected from photographs taken at sea but also from museum specimens and beached individuals. Whereas molt duration generally increased with species’ size, molt duration in Parasitic and Pomarine jaegers was surprisingly similar given their size difference. Larger species started primary molt earlier and showed more overlap with postbreeding migration, such that there was complete overlap in Great Skua but no overlap in Long-tailed Jaeger. Within jaeger species, the first primary molt cycle took longer than later molt cycles. We suggest that, unlike birds in their first primary molt cycle, birds in their second or subsequent primary molt cycles are time-constrained to complete primary molt before the onset of prebreeding long-distance migration. By contrast, molt duration did not differ between age classes of Great Skuas. Adult Great Skuas may have overcome the time constraint by completely overlapping molt and postbreeding migration. Molt-migration overlap is generally rare in birds but may be feasible for Great Skuas given their shorter migration distance and low migration speed.

Genetic transformation of microalgae remains a challenge due to poor intracellular delivery of exogenous molecules. This limitation is caused by the structure and composition of the cell wall and cell membrane of each species. Moreover, successful delivery of proteins or nucleic acids cannot be assessed by determining transformability since their functionality is not always known in the studied microorganism. We propose a quick and effective screening tool for the prediction and optimization of electroporation settings by monitoring cell permeability and viability using Sytox Green and propidium iodide respectively. We determined voltage settings for the microalgae Chlamydomonas reinhardtii, Chlorella vulgaris, Neochloris oleoabundans and Acutodesmus obliquus. To evaluate the predicted settings, we delivered labelled DNA and proteins into the cells. We demonstrated that high transformation efficiencies can be accomplished when predicted values were applied with functional plasmids. Additionally, we increased transformation efficiencies by testing cell concentrations, light intensities and fragment sizes. This method can be used to determine suitable transformation conditions for non-transformed microalgae species and to increase the insight on established transformation protocols.

A role for TLR10 in obesity and adipose tissue morphology

Toll like receptors (TLRs) are expressed in adipose tissue and promote adipose tissue inflammation during obesity. Recently, anti-inflammatory properties have been attributed to TLR10 in myeloid cells, the only member of the TLR family with inhibitory activity. In order to assess whether TLR10-induced inhibition of inflammation may be protective during the development of obesity and metabolic abnormalities we used transgenic human TLR10 mice (hTLR10tg) and wild type (WT) controls on a C57B6J background. HFD-feeding enhanced TLR10 expression in the adipose tissue, and HFD-fed hTLR10tg mice displayed reduced adipocyte size, adipose tissue weight, and a trend toward lower plasma insulin levels compared to WT mice. In humans, obese individuals with polymorphisms in the TLR10 gene displayed reduced macrophage infiltration in the adipose tissue accompanied by a trend to lower leptin levels and higher adiponectin levels in plasma. In healthy individuals with the same polymorphisms in the TLR10 gene we did not observe any difference in plasma concentrations of leptin and adiponectin. We conclude that TLR10 impacts adipose tissue morphology in obesity. Larger studies in humans are warranted to assess its potential value as therapeutic target in metabolic syndrome and type 2 diabetes.
Neochloris oleoabundans is worth its salt : Transcriptomic analysis under salt and nitrogen stress

Neochloris oleoabundans is an oleaginous microalgal species that can be cultivated in fresh water as well as salt water. Using salt water gives the opportunity to reduce production costs and the fresh water footprint for large scale cultivation. Production of triacylglycerols (TAG) usually includes a biomass growth phase in nitrogen-replete conditions followed by a TAG accumulation phase under nitrogen-deplete conditions. This is the first report that provides insight in the saline resistance mechanism of a fresh water oleaginous microalgae. To better understand the osmoregulatory mechanism of N. oleoabundans during growth and TAG accumulating conditions, the transcriptome was sequenced under four different conditions: fresh water nitrogen-replete and -deplete conditions, and salt water (525 mM dissolved salts, 448mM extra NaCl) nitrogen-replete and -deplete conditions. In this study, several pathways are identified to be responsible for salt water adaptation of N. oleoabundans under both nitrogen-replete and -deplete conditions. Proline and the ascorbate-glutathione cycle seem to be of importance for successful osmoregulation in N. oleoabundans. Genes involved in Proline biosynthesis were found to be upregulated in salt water. This was supported by Nuclear magnetic resonance (NMR) spectroscopy, which indicated an increase in proline content in the salt water nitrogen-replete condition. Additionally, the lipid accumulation pathway was studied to gain insight in the gene regulation in the first 24 hours after nitrogen was depleted. Oil accumulation is increased under nitrogen-deplete conditions in a comparable way in both fresh and salt water. The mechanism behind the biosynthesis of compatible osmolytes can be used to improve N. oleoabundans and other industrially relevant microalgal strains to create a more robust and sustainable production platform for microalgae derived products in the future.

Chlamydomonas reinhardtii - Enhanced stearic acid - RNA silencing

In this study, stearoyl-ACP desaturase (SAD), the enzyme that converts stearic acid into oleic acid, is silenced by artificial microRNA in the green microalga Chlamydomonas reinhardtii. Two different constructs, which target different positions on the mRNA of stearoyl-ACP desaturase, were tested. The mRNA levels for SAD were reduced after the silencing construct was induced. In one of the strains, the reduction in SAD mRNA resulted in a doubling of the stearic acid content in triacylglycerol molecules, which shows that stearic acid production in microalgae is possible.

The microalgae Tetradesmus obliquus is able to maintain a high photosynthetic efficiency under nitrogen limitation and is considered a promising green microalgae for sustainable production of diverse compounds, including biofuels. Here, we report the first draft whole-genome shotgun sequencing of T. obliquus. The final assembly comprises 108,715,903 bp with over 1,368 scaffolds.

The first draft genome of Scenedesmus obliquus (UTEX 393). The assembly yielded 9191 contigs (min length 200bp) for a total length of 96197736 bp. The scaffolding yielded 1680 sequences for a total of 126848291 bp. We were not able to recover four chromosomes and the two organelle genomes. Few scaffolds were found to cover 99.97% of the reference chloroplast genome. The mitochondrial genome was too fragmented and the scaffolds covered only 46% of UTEX78 and KS3-2.

Comment les plantes poussent-elles ? Le nombre d’or est-il omniprésent dans leur architecture ? Leurs structures sont-elles fractales ? Ont-elles un langage ? Une grammaire ? Il y a du vrai dans ces questions, mais il faut aller plus loin. Il faut identifier, modéliser et simuler le rôle des organes d’une plante dans sa croissance et leur fonctionnement.

La modélisation des plantes fait l’objet de recherches à l’interface de disciplines biologiques (botanique, agronomie, génétique, écophysiologie) des mathématiques appliquées et de l’informatique. Elle permet de créer les modèles de développement de l’architecture des plantes, mais aussi les modèles de production végétale, expression de la croissance des organes.

Les auteurs de cet ouvrage nous exposent les fondements biologiques, mathématiques et informatiques qui permettent d’exprimer le fonctionnement des bourgeons, la production photosynthétique de la biomasse et sa répartition dans les organes d’une plante. La simulation de la croissance des plantes devient alors possible sous la forme de modèles dynamiques et sa représentation, sous la forme d’images de synthèse. Ces modèles autorisent de multiples applications. De nombreuses plantes sont présentées (herbacées, arbustes et arbres) ; leur modélisation a une visée pratique en agriculture, en gestion des ressources naturelles et de l’environnement et en représentation des paysages.

Rédigé par des botanistes, des agronomes, des mathématiciens et des informaticiens, cet ouvrage collectif est le fruit de quarante années de recherches conduites par Philippe de Reffye et ses collègues, avec des collaborations scientifiques en France, en Hollande, en Chine et en Afrique. Il s’adresse aux chercheurs, enseignants et étudiants en biologie, en agronomie et en sciences de la vie, aux architectes paysagistes, écologues, qui s’intéressent à la croissance des plantes, à leur modélisation et à leur représentation. Il s’adresse aussi à toutes les communautés des sciences exactes (mathématiciens, informaticiens, physiciens) intéressées par la simulation du vivant.

How effective is road mitigation at reducing road-kill? A meta-analysis

Road traffic kills hundreds of millions of animals every year, posing a critical threat to the populations of many species. To address this problem there are more than forty types of road mitigation measures available that aim to reduce wildlife mortality on roads (road-kill). For road planners, deciding on what mitigation method to use has been problematic because there is little good information about the relative effectiveness of these measures in reducing road-kill, and the costs of these measures vary greatly. We conducted a metaanalysis using data from 50 studies that quantified the relationship between road-kill and a mitigation measure designed to reduce road-kill. Overall, mitigation measures reduce roadkill by 40% compared to controls. Fences, with or without crossing structures, reduce roadkill by 54%. We found no detectable effect on road-kill of crossing structures without fencing. We found that comparatively expensive mitigation measures reduce large mammal road-kill much more than inexpensive measures. For example, the combination of fencing and crossing structures led to an 83% reduction in road-kill of large mammals, compared to a 57% reduction for animal detection systems, and only a 1% for wildlife reflectors. We suggest that inexpensive measures such as reflectors should not be used until and unless their effectiveness is tested using a high-quality experimental approach. Our meta-analysis also highlights the fact that there are insufficient data to answer many of the most pressing questions that road planners ask about the effectiveness of road mitigation measures, such as whether other less common mitigation measures (e.g., measures to reduce traffic volume and/or speed) reduce road mortality, or to what extent the attributes of crossing structures and fences influence their effectiveness. To improve evaluations of mitigation effectiveness, studies should incorporate data collection before the mitigation is applied, and we recommend a minimum study duration of four years for Before-After, and a minimum of either four years or four sites for Before-After-Control-Impact designs.
