    Bridging Bacteria and the Gut : Functional Aspects of Type IV Pili

    Cell-surface-located proteinaceous appendages, such as flagella and fimbriae or pili, are ubiquitous in bacterial communities. Here, we focus on conserved type IV pili (T4P) produced by bacteria in the intestinal tract, one of the most densely populated human ecosystems. Computational analysis revealed that approximately 30% of known intestinal bacteria are predicted to produce T4P. To rationalize how T4P allow intestinal bacteria to interact with their environment, other microbiota members, and host cells, we review their established role in gut commensals and pathogens with respect to adherence, motility, and biofilm formation, as well as protein secretion and DNA uptake. This work indicates that T4P are widely spread among the known members of the intestinal microbiota and that their contribution to human health might be underestimated.

    Purpose: The environmental impact, resource use and waste generation of packaging has been a topic of worldwide debate. This resulted in founding the Netherlands Institute for Sustainable Packaging (KIDV), which aims to facilitate the reduction of these impacts. Within KIDV’s scientific programme, an LCA-based tool was developed to show packaging design students the underlying causes of this impact. Researchers can assess packaging chain alternatives with the tool, which is presented in the first part of the paper. Methods: The LCA-based tool, EnvPack, encompassed three consumer products: non-carbonated beverage, shower gel and ready-to-eat soup. Each product had three to four different packaging alternatives. The packaging cradle-to-grave life cycles were defined in terms of materials and processes and included detailed parametrisation of the end-of-life. Packaging-related product losses have been included in EnvPack. For the impact assessment of the product-packaging combinations, four methods were included, each with a different perspective. These were a modified ReCiPe midpoint method, ReCiPe endpoint, cumulative energy demand and a Circular Economy method based on ReCiPe. Packaging for material analysis was collected at Dutch supermarkets. For establishing packaging-related product losses, explorative measurements were made. Microsoft Excel was used to construct EnvPack. Results and discussion: Researchers and design students can select up to four different packaging alternatives per product, including one self-designed packaging. Packaging-related product losses can be included or not in the assessment. For the beverage, an out-of-home consumption situation can be selected, which affects the end-of-life of the packaging. The contribution of several life cycle stages and of impact categories are presented as graphs for the design students; detailed tables are available for researchers. The tool compares two assessment methods at a time. The effect of different methods on the ranking of the packaging alternatives is a topic of the second part paper. Conclusions: In comparison with existing LCA-based packaging tools, EnvPack includes four different assessment methods that all offer a single score comparison of alternatives. EnvPack is freely available for participating Dutch universities.

    Consumer response to packaging design : The role of packaging materials and graphics in sustainability perceptions and product evaluations

    Building on theories of cue utilization, this paper investigates whether and how packaging sustainability influences consumer perceptions, inferences and attitudes towards packaged products. A framework is tested in an empirical study among 249 students using soup products varying in packaging material and graphics. The findings show that (packaging) sustainability is a highly salient association but is only moderately important for consumer attitudes. A comparison between consumer judgments and life-cycle assessment indicates that consumers rely on misleading, inaccurate lay beliefs to judge packaging sustainability and are therefore susceptible to making ineffective environmental decisions. The research also demonstrates the power of packaging in shaping perceptions of food products. Particularly, it shows that changes in actual environmental impacts (by altering packaging materials) affect not only sustainability perceptions but also several other benefits, such as perceived taste and quality. At the same time, consumers' sustainability assessments are also highly influenced by mere graphical packaging cues that have no obvious actual sustainability consequences.

    In many delta areas hydraulic structures are key elements in water management strategies for fresh water supply and flood risk management. Adaptation of delta areas to changing climatological and societal conditions will be in pace with the renovation and replacement of these hydraulic structures. Since hydraulic structures are prone to deterioration, their performance diminishes over time. Changes in society, the economy, and the physical environment can also alter the functionality of structures, or have an impact on their performance. Although faced with deterioration and exogenous changes, timing of replacement is essential because replacing too early leads to insufficient use of invested capital, while replacing too late leads to loss of societal benefits. This article explores the timing of replacement using adaptation tipping points. We indicate three drivers – deterioration, biophysical change, and socio-economic change – that determine the moment in time when replacement becomes necessary. Moreover, we conclude that for determining the moment of replacement, at the very least, the objectives, maintenance and operations of hydraulic structures need to be taken into account. This exploration is illustrated with the task of replacing seven hydraulic structures in the River Meuse.

    Seed is the physical embodiment of the invention of the plant breeder. Plant varieties thus constitute a special form of innovation, and an assessment of intellectual property right (IPR) systems needs to take this into account. This thesis concentrates on IPRs but breeders do have a number of means by which they can capture part of the benefits from the cultivation of their new variety, rather than these falling into to public domain.

    Background, aim and scope - The environmental impact of building products made from heavy metals has been a topic of discussion for some years. This was fuelled by results of life cycle assessments (LCAs), where the emission of heavy metals strongly effected the results. An issue was that the characterisation factors of the Centre for Environmental Studies (CML) 2000 life cycle impact assessment (LCIA) methodology put too much emphasis on the impact of metal emissions. We adjusted Zn characterisation factors according to the most recent insights in the ecotoxicity of zinc and applied them in an LCA using zinc gutters and downpipes as an example. Materials and methods - The CML 2000 methodology was used to assess the environmental impact of the zinc products. To adjust the Zn characterisation factors, the uniform system for the evaluation of substances (USES)–LCA model and the biotic ligand model were used. Results and discussion - The first correction was based on updating the effect values for zinc. This resulted in a reduction of the characterisation factors for zinc to 42% of their original values. Additional correcting for the bioavailability of zinc leads to final Zn characterisation factors for the freshwater aquatic ecotoxicity potential (FAETP), the marine aquatic ecotoxicity potential (MAETP) and the terrestrial ecotoxicity potential (TETP) of 25%, 42% and 0.006%, respectively, of the original values. The CML 2000 LCIA methodology is based on the predicted no-effect concentration (PNEC) of a substance. PNEC is not value-free as political considerations are used to decide on it. Using a more robust toxicity measure as the hazardous concentration at which 50% of the species is affected (HC50) will provide value-free results. The production of standard high-grade zinc shows main contributions to six of the ten environmental impact categories. The recycling of zinc at the end of the life cycle shows beneficial effects for these same categories. Despite the reduction of the characterisation factor of Zn, the runoff emissions of Zn are still dominant. Conclusions and recommendations - To improve LCA characterisation factors for ecotoxicity in the CML 2000 methodology, it is recommended to use either the geometric mean of the effect data or the HC50. The HC50 should be based upon the EC50 values from chronic ecotoxicity tests. It is proposed to include the bioavailability of metals in LCA in three steps: (1) separate soluble fraction, (2) separate dissolved fraction and (3) separate bioavailable fraction. The issue of essentiality could not be resolved in this study. However, this could be accounted for by leaving out the fraction of the emission below the maximum permissible admission

    A 3-ureidobenzo-1,2,4-triazine 1-N-oxide (1) was synthesized successfully. The derivative displays an acceptor-donor-acceptor-acceptor (ADAA) hydrogen-bonding motif in CDCl3 and DMSO-d6 solution as well as in the solid state. Although moderately strong association of 1 was observed with DAD motifs, nonspecific binding is observed with ureidopyridines featuring a complementary DADD array. Density functional calculations of conformations 1a and 1b together with two complexes revealed the clearly nonplanar geometry of the multiply hydrogen-bonded complex, in which some bonds are significantly longer (3.2 Å) than is optimal for H-bonds. As a result, only very small free energies of association were calculated, in line with the experimentally observed absence of specific assembly of the components.

    This paper describes the contours of a Dutch monitoring and research site for climate change and related atmospheric processes. The station has large benefits for atmospheric science, both in The Netherlands and internationally. It provides a platform for collaboration in this important field, and will provide the routine observations needed to assess the impact of the different atmospheric parameters on the local climate. The station fits in directly in the selected group of global monitoring networks that are currently operational or being set up to address the problems of climate. In addition, the station can play a major role in supporting worldwide satellite measurements of climate related parameters. The only way to get a global picture of the essential climate change parameters can be found in the combination of satellite measurements and ground-based stations equipped with advanced remote sensing and in situ instrumentation. Furthermore, the combined expertise of European universities and research institutes, encompassing the whole field of atmospheric research, offers a unique chance for the training of young scientists. The research site is an attractive center for international young scientists to develop and deepen their skills

    Vanaf 2003 werkt het Milieu- en Natuurplanbureau (MNP) aan de ontwikkeling van een methodiek (indicatoren en analysekader) om de kosteneffectiviteit van het natuurbeleid te bepalen. Achtergrond van deze studie vormt de wens van de ministeries van Financien en Landbouw, Natuur en Voedselkwaliteit om het inzicht in de relatie tussen de kosten van het natuurbeleid en de doelbereiking te vergroten. Deze methodiek moet het in de toekomst mogelijk maken om de vraag te beantwoorden of de doelen van het huidige natuurbeleid kunnen worden gerealiseerd met minder inzet van financiële middelen of met een gelijke inzet van middelen een groter doelbereik gehaald kan worden. De methodiekontwikkeling richt zich primair op de realisatie van de Ecologische Hoofdstructuur (EHS) zowel qua oppervlakte als in kwalitatieve zin. De ontwikkeling van een kosteneffectviteits-methodiek is een uitdaging van formaat, vanwege de complexiteit van het natuurbeleid, het multidisciplinaire karakter ervan en de onoverkomelijkheid tal van keuzes te maken die zowel wetenschappelijk gezien als bestuurlijk voor discussie vatbaar zijn. Dit rapport moet dan ook worden beschouwd als een tussenrapport waarin de keuzes die tot nu toe zijn gemaakt met betrekking tot de indicatoren voor doelbereiking, kosten en kosteneffectiviteit uitvoerig worden beschreven en onderbouwd. Behalve een presentatie van de methodiek, staan in deze tussenrapportage de resultaten van een kosteneffectviteitsanalyse van twee voorbeeldstudies, namelijk voor de natuurdoelen van heide en laagveenmoeras.     In het kader van het onderzoeksprogramma 'Biologische productiesystemen in de akkerbouw en vollegrondsgroenteteelt' is de afgelopen vier jaar onderzoek gedaan naar de mogelijkheden die veredeling kan bieden om het wortelstelsel van uien te verbeteren. Dit lijkt mogelijk door specifieke soortkruisingen met stengelui (Japanse bladui) en Allium roylei, met als bijkomend voordeel dat naast de selectie op een beter type wortelstelsel ook geselecteerd kan worden op resistentie tegen de bladschimmels valse meeldauw en bladvlekkenziekte. Een opvallend onderzoeksresultaat was het effect van arbusculaire mycorrhiza (AM) op het verbeteren van het wortelstelsel en de productiviteit van stengelui, en mogelijk ook ui

    In verschillende gremia binnen het Ministerie van Verkeer en Waterstaat en daarbuiten is vastgesteld dat er behoefte is aan verbreding van het gegevensaanbod om de beleidsvoering te ondersteunen. Met de bestaande monitoringprogramma's worden voornamelijk fysische, chemische en biologische gegevens verzameld. Deze dienen te worden aangevuld met gegevens over de zogenaamde gamma-aspecten van het waterbeheer. De projectgroep GGB heeft drie terreinen onderscheiden waarop de nieuwe informatiebehoefte zichricht, namelijk 'geld', 'gevoel' en 'beleidsprocessen'. De projectgroep beoogt voor deze drie terreinen onderbouwd in beeld te brengen aan welk type informatie binnen Verkeer en Waterstaat behoefte bestaat. Daartoe zijn eerst deze drie terreinen nader gedefinieerd en is de informatiebehoefte met betrekking tot deze drie terreinen globaal verkend (deelrapport 780.1). Behalve deze watvraag is de vraag aan de orde gesteld of de gesignaleerde specifieke informatiebehoefte zich wel leent voor het type gegevensverzameling dat we duiden als klassieke monitoring. In het vervolgonderzoek, (deelrapport 780.2) is aan de hand van het voorbeeld de toepassing van de watertoets de specifieke informatiebehoefte rond monitoring van beleidsprestaties(beleidsevaluatie) nader uitgewerkt. Voor het voorbeeld diffuse bronnen zijn de specifieke informatiebehoefte rond monitoring van de omgeving van het beleidsproces en veranderingen hierin geduid. Ook in dit deelonderzoek is nagegaan in welke mate via klassieke monitoring inde gesignaleerde informatiebehoefte kan worden voorzien.
