    Tebuconazole (TEB) is a widely used triazole fungicide, but the toxicokinetics of its human metabolites are not fully described. For proper interpretation of biological monitoring data, knowledge on the metabolism and elimination of the compound is required. A human volunteer study was performed with the aim to describe the time courses of urinary excretion after controlled oral and dermal administration of TEB. Six healthy volunteers (three males and three females) received on separate occasions a single oral dose of 1.5 mg of TEB and a single dermal dose of 2.5 mg during 1 h. In addition to a pre-exposure urine sample, complete urine voids were collected over 48 h post-administration. The main metabolite hydroxy-tebuconazole (TEB-OH) was quantified in each urine sample. Peak excretion rates after oral and dermal administration were reached after 1.4 and 21 h, mean elimination half-lives were 7.8 and 16 h, and recoveries within 48 h were 38% and 1%, respectively. The time courses of excretion were compared to simulations with an established physiologically based toxicokinetic model for TEB that was extended with a parallel model for TEB-OH. Overall, TEB-OH was rapidly excreted into urine after oral exposure, and renal elimination was considerably slower after dermal exposure. Urinary time courses between individuals were similar. The model predictions were in good agreement with the observed time courses of excretion.

    The t(8;21) acute myeloid leukemia (AML)-associated oncoprotein AML1-ETO disrupts normal hematopoietic differentiation. Here, we have investigated its effects on the transcriptome and epigenome in t(8,21) patient cells. AML1-ETO binding was found at promoter regions of active genes with high levels of histone acetylation but also at distal elements characterized by low acetylation levels and binding of the hematopoietic transcription factors LYL1 and LMO2. In contrast, ERG, FLI1, TAL1, and RUNX1 bind at all AML1-ETO-occupied regulatory regions, including those of the AML1-ETO gene itself, suggesting their involvement in regulating AML1-ETO expression levels. While expression of AML1-ETO in myeloid differentiated induced pluripotent stem cells (iPSCs) induces leukemic characteristics, overexpression increases cell death. We find that expression of wild-type transcription factors RUNX1 and ERG in AML is required to prevent this oncogene overexpression. Together our results show that the interplay of the epigenome and transcription factors prevents apoptosis in t(8;21) AML cells.

    Background Elderly people may benefit from sensory stimulation to increase food intake since anorexia of ageing is prevalent among them. An optimal MSG concentration may increase the palatability of foods but this depends on the food and chemosensory status of the taster. Currently, the results on taste enhancing to increase intake are inconsistent. Objective To find an optimal preferred MSG concentration in mashed potatoes, spinach and ground beef and to determine whether this concentration increases consumption of these foods among institutionalized elderly people. Design Single blind within subject cross-over study performed at the laboratory and in the residents’ own apartments. Participants 33 elderly and 29 young people in the sensory study and 53 elderly people in the intake study. Measurements Pleasantness of the foods was rated of the foods each with 0, 0.5, 0.8, 1.3 and 2.0 g of MSG/100g. Intake was measured by weighing back leftovers of 2 meals with MSG (0.5% in mashed potatoes, 2% in spinach and ground meat) and without MSG. Results 0.5% MSG (p0.68). Conclusion MSG (0.5% and 2%) does not guarantee a higher intake among elderly. The chemosensory heterogeneity of the elderly population requires more individual flavor enhancement to improve the dietary intake and sensory experience

    In artikel 27 Wet OB 1968 is de landbouwregeling uitgewerkt. Landbouwers, veehouders, tuinbouwers en bosbouwers zijn over prestaties genoemd in dit artikel geen omzetbelasting verschuldigd, maar hebben ook geen recht op aftrek van voorbelasting. De ondernemer kan opteren om in de heffing van de omzetbelasting te worden betrokken als een normale ondernemer. In deze evaluatie wordt onderzocht of de landbouwregeling op een doeltreffende (effectieve) en doelmatige (efficiënte) manier leidt tot het bereiken van de geformuleerde doelstelling.     In dit onderzoek hebben we via een enquête en interviews onder agrarische natuurverenigingen (ANV’s) gekeken naar de mogelijkheden om de effectiviteit van het agrarisch natuurbeheer te versterken. Opvallende uitkomst is dat ANV’s die een collectief contract hebben afgesloten voor weidevogel- en slootkantbeheer, meer directe en indirecte maatregelen nemen om die effectiviteit te vergroten. Directe maatregelen betreffen bijvoorbeeld zogenaamde ecologische sturing, zoals het afsluiten met leden van contracten die ecologisch gezien op de juiste plaats liggen. Indirecte maatregelen betreffen het geven van een (al dan niet verplichte) cursus agrarisch natuurbeheer of een schouwcommissie, die beoordeelt of het beheer effectief is geweest. We bevelen de provincies dan ook aan om meer collectieve contracten mogelijk te maken, zoals voor landschapselementen.     Thermal degradation of individual glucosinolates within the plant matrix was studied. Red cabbage samples were heated at different temperatures for various times. To rule out the influence of enzymatic breakdown and to focus entirely on the thermal degradation of glucosinolates, myrosinase was inactivated prior to the thermal treatments. All identified glucosinolates degradation when heated at temperatures above 100 °C. The indole glucosinolates 4-hydroxy-glucobrassicin and 4-methoxyglucobrassicin showed the highest degree of showed degradation, even at temperatures below 100 °C. Kinetic parameters have been estimated for the degradation that could be described by first-order kinetics. At temperatures below 110 °C indole glucosinolates have a significant higher degradation rate constant as compared to aliphatic glucosinolates. The breakdown of 4-hydroxyglucobrassicin seems to consist of two parallel reaction pathways. Based on the proposed degradation kinetics and the estimated parameters, the degree of thermal degradation of all individual glucosinolates at standardized heating conditions (blanching, cooking and canning) was simulated. Glucosinolates are expected to be not very susceptible to thermal degradation during blanching conditions. Cooking will cause more thermal degradation to indole glucosinolates (38%) as compared to aliphatic glucosinolates (8%). Canning, the most severe heat treatment, will result in significant thermal degradation (73%) of the total amount of glucosinolates
