    In arid and semi-arid ecosystems, shortage of water can trigger changes in landscapes’ structures and function leading to degradation and desertification. Hydrological connectivity is a useful framework for understanding water redistribution and scaling issues associated with runoff and sediment production, since human and/or natural disturbances alter surface water availability and pathways increasing/decreasing connectivity. In this paper, we illustrate the use of the connectivity framework for several examples of dryland systems that are analysed at a variety of spatial and temporal scales. In doing so, we draw particular attention to the analysis of coevolution of system structures and function, and how they may drive threshold behaviour leading to desertification and degradation. We first analyse the case of semi-arid rangelands, where feedbacks between the decline in vegetation density and landscape erosion reinforces degradation processes driven by changes in connectivity until a threshold is crossed above which the return to a functional system is unlikely. We then focus on semi-arid wetlands, where decreases in water volumes promote terrestrial vegetation encroachment that changes drainage conditions and connectivity, potentially reinforcing redistribution of flow paths to other wetland areas. The analysis of dryland wetlands is based on a novel hydrologic connectivity index derived using inundation requirements for wetland vegetation associations. The examples presented highlight the need to incorporate a coevolutionary framework for the analysis of changing connectivity patterns and the emergence of thresholds in arid and semi-arid systems.

    Millet is a species of the grass family used in cattle pastures, for hay, for silage, and in soil cover in no-tillage systems. However, studies on seed production and post-harvest techniques for this species have not been sufficiently developed, considering the demand for and potential of this crop. The objective of this study was to monitor the physiological quality of millet seeds stored in porous packaging. Millet seeds were stored under four ambient conditions (natural laboratory environment, dry chamber, refrigerator, and freezer). Physiological quality was evaluated by germination and vigor tests before and during storage each year, up to six years for the dry chamber and natural laboratory environment, and up to 12 years for the refrigerator and freezer. Analysis of variance was conducted on the data, which were in 4 × 6 and 2 × 12 (environment and storage period) factorial arrangements. The means of the environments were compared by the Tukey test (p ≤ 0.05) and the storage periods by regression analysis. The germination capacity of millet seeds is best conserved in a refrigerator and freezer, and they remain viable for up to twelve years of storage under these conditions.

    A 2-year field experiment was conducted to determine the effects of Fusarium sporotrichioides, F. langsethiae, or F. poae on durum wheat plants artificially inoculated at different growth stages. The percentage of symptomatic kernels was similar among the three species, but incidence of infected kernels was lower for F. langsethiae. Kernel colonization was higher when plants were inoculated before and during anthesis for F. sporotrichioides and F. poae, but unaffected by timing of inoculation for F. langsethiae. Production of T-2/HT-2 toxins was higher for F. sporotrichioides than for F. langsethiae. Significant accumulations of nivalenol were detected for F. poae. Across all three species, there was a weak correlation (r = 0.16; P = 0.031) between the incidences of symptomatic and infected kernels, but a stronger correlation (r = 0.53; P < 0.001) between infection incidence and the quantity of fungal DNA (species-specific) in kernels. Mycotoxin content was correlated (r > 0.58; P < 0.007) with infection incidence or fungal DNA in kernels, but only for F. sporotrichioides and F. poae.

    Adoption of innovation in agriculture: a critical review of economic and psychological models

    Two main models have been used to analyse farmers’ decisions to
    adopt an innovation; the first is based on the concept of utility maximisation
    (UM) and the second is based on the theory of planned behaviour (TPB). This
    study uses a vote-count method to identify the effect of different variables on
    farmers’ adoption decisions in 36 studies using either UM or the TPB. Results
    from the UM studies show that the explanatory variables mostly have an
    insignificant effect on the adoption decision. When the effects are significant,
    the sign of the effect is inconsistent across studies. Results from the TPB
    studies show that correlations between the psychological constructs used in this
    type of model are significant in most cases. However, most variables are only
    used in one or two studies and it is therefore not

    Mycopathologia was founded in 1938 to ‘diffuse the understanding of fungal diseases in man and animals among mycologists.’ This was an important mission considering that pathogenic fungi for humans and animals represent a tiny minority of the estimated 1.5–5 million fungal inhabitants on Earth. These pathogens have diverged from the usual saprotrophic lifestyles of most fungi to colonize and infect humans and animals. Medical and veterinary mycology is the subdiscipline of microbiology that dwells into the mysteries of parasitic, fungal lifestyles. Among the oldest continuing scientific publications on the subject, Mycopathologia had its share of ‘classic papers’ since the first issue was published in 1938. An analysis of the eight decades of notable contributions reveals many facets of host–pathogen interactions among 183 volumes comprising about 6885 articles. We have analyzed the impact and relevance of this body of work using a combination of citation tools (Google Scholar and Scopus) since no single citation metric gives an inclusive perspective. Among the highly cited Mycopathologia publications, those on experimental mycology accounted for the major part of the articles (36%), followed by diagnostic mycology (16%), ecology and epidemiology (15%), clinical mycology (14%), taxonomy and classification (10%), and veterinary mycology (9%). The first classic publication, collecting nearly 200 citations, appeared in 1957, while two articles published in 2010 received nearly 150 citations each, which is notable for a journal covering a highly specialized field of study. An empirical analysis of the publication trends suggests continuing interests in novel diagnostics, fungal pathogenesis, review of clinical diseases especially with relevance to the laboratory scientists, taxonomy and classification of fungal pathogens, fungal infections and carriage in pets and wildlife, and changing ecology and epidemiology of fungal diseases around the globe. We anticipate that emerging and re-emerging fungal pathogens will continue to cause significant health burden in the coming decades. It remains vital that scientists and physicians continue to collaborate by learning each other’s language for the study of fungal diseases, and Mycopathologia will strive to be their partner in this increasingly important endeavor to its 100th anniversary in 2038 and beyond.
    microRNA 193a-5p Regulates Levels of Nucleolar- and Spindle-associated Protein 1 to Suppress Hepatocarcinogenesis

    Background & Aims: We performed an integrated analysis to identify microRNAs (miRNAs) and mRNAs with altered expression in liver tumors from 3 mouse models of hepatocellular carcinoma (HCC) and human tumor tissues. Methods: We analyzed miRNA and mRNA expression profiles of liver tissues from mice with diethylnitrosamine-induced hepatocarcinogenesis, conditional expression of lymphotoxin alpha and lymphotoxin beta , or inducible expression of a Myc transgene (Tet-O-Myc mice), as well as male C57BL/6 mice (controls). miRNA mimics were expressed and miRNAs and mRNAs were knocked down in human (Huh7, Hep3B, JHH2) hepatoma cell lines; cells were analyzed for viability, proliferation, apoptosis, migration, and invasion. Cells were grown as xenograft tumors in nude mice and analyzed. We combined in-silico target gene prediction with mRNA profiles from all 3 mouse models. We quantified miRNA levels in 146 fresh-frozen tissues from patients (125 HCCs, 17 matched non-tumor tissues, and 4 liver samples from patients without cancer) and published human data sets and tested correlations with patient survival times using Kaplan-Meier curves and the log-rank test. Levels of NUSAP1 mRNA were quantified in 237 HCCs and 5 non-tumor liver samples using the Taqman assay. Results: Levels of the microRNA 193a-5p (MIR193A-5p) were reduced in liver tumors from all 3 mouse tumor models and in human HCC samples, compared with non-tumor liver tissues. Expression of a MIR193A-5p mimic in hepatoma cells reduced proliferation, survival, migration, and invasion and their growth as xenograft tumors in nude mice. We found nucleolar and spindle associated protein 1 (NUSAP1) to be a target of MIR193A-5p; HCC cells and tissues with low levels of MIR193A-5p had increased expression of NUSAP1. Increased levels of NUSAP1 in HCC samples correlated with shorter survival times of patients. Knockdown of NUSAP1 in Huh7 cells reduced proliferation, survival, migration, and growth as xenograft tumors in nude mice. Hydrodynamic tail-vein injections of a small hairpin RNA against NUSAP1 reduced growth of AKT1-MYC–induced tumors in mice. Conclusions: MIR193A-5p appears to prevent liver tumorigenesis by reducing levels of NUSAP1. Levels of MIR193A-5p are reduced in mouse and human HCC cells and tissues, leading to increased levels of NUSAP1, associated with shorter survival times of patients. Integrated analyses of miRNAs and mRNAs in tumors from mouse models can lead to identification of therapeutic targets in humans.
    Comparison of three modelling approaches for predicting deoxynivalenol contamination in winter wheat

    Forecasting models for mycotoxins in cereal grains during cultivation are useful for pre-harvest and post-harvest mycotoxin management. Some of such models for deoxynivalenol (DON) in wheat, using two different modelling techniques, have been published. This study aimed to compare and cross-validate three different modelling approaches for predicting DON in winter wheat using data from the Netherlands as a case study. To this end, a published empirical model was updated with a new mixed effect logistic regression method. A mechanistic model for wheat in Italy was adapted to the Dutch situation. A new Bayesian network model was developed to predict DON in wheat. In developing the three models, the same dataset was used, including agronomic and weather data, as well as DON concentrations of individual samples in the Netherlands over the years 2001–2013 (625 records). Similar data from 2015 and 2016 (86 records) were used for external independent validation. The results showed that all three modelling approaches provided good accuracy in predicting DON in wheat in the Netherlands. The empirical model showed the highest accuracy (88%). However, this model is highly location and data-dependent, and can only be run if all of the input data are available. The mechanistic model provided 80% accuracy. This model is easier to implement in new areas given similar mycotoxin-producing fungal populations. The Bayesian network model provided 86% accuracy. Compared with the other two models, this model is easier to implement when input data are incomplete. In future research, the three modelling approaches could be integrated to even better support decision-making in mycotoxin management.
