    Oomycetes form a deep lineage of eukaryotic organisms that includes a large number of plant pathogens that threaten natural and managed ecosystems. We undertook a survey to query the community for their ranking of plant pathogenic oomycete species based on scientific and economic importance. In total, we received 263 votes from 62 scientists in 15 countries for a total of 33 species. The Top 10 species and their ranking are: (1) Phytophthora infestans; (2, tied) Hyaloperonospora arabidopsidis; (2, tied) Phytophthora ramorum; (4) Phytophthora sojae; (5) Phytophthora capsici; (6) Plasmopara viticola; (7) Phytophthora cinnamomi; (8, tied) Phytophthora parasitica; (8, tied) Pythium ultimum; and (10) Albugo candida. The article provides an introduction to these 10 taxa and a snapshot of current research. We hope that the list will serve as a benchmark for future trends in oomycete research.

    The market for greenhouse tomatoes requires a production system with lower use and risks of pesticides. These requirements culminate in the tight restrictions on residues for German retailers, both in number and in levels of residues. Germany is an important import country for tomatoes because of the size of the German market. Furthermore, exports to Germany entail a price premium. For that reason the revenues of IPM in greenhouse tomatoes are more important than the costs. Consequently, the experiments in task 3 and the ex post assessment in task 4 of work package 7 of the Pure programme should focus on IPM strat egies by which the German residue restrictions can be respected.
    ENDURE Speeds Development of Biological

    Research conducted under the European Commission- funded ENDURE Network of Excellence (2007– 10) has led to the development of a new approach aimed at the fast and cost-effective development of biological control products. Called SelectBioControl, the approach is described as encompassing all the successive steps crucial to the development, production, registration and marketing of such products. The development of SelectBioControl was led by Jürgen Köhl, from Plant Research International (PRI), part of Wageningen University and Research Centre (the Netherlands), working in cooperation with ENDURE colleagues from France's National Institute for Agricultural Research (INRA), Italy's National Research Council (CNR) and the biocontrol industry body, the International Biocontrol Manufacturers' Association (IBMA). ........     The review of published scientific literature on the biological control of selected pests and diseases has lead to the identification of clear knowledge gaps highlighted in previous chapters. Further bottlenecks were revealed by seeking the possible reasons for the striking discrepancy between the rich inventory of potential biocontrol agents described by scientists and a very small number of commercial products on the market. The unique diversity of expertise and concerns allowed the group to set up very complementary approaches to tackle the issue of the factors of success of biocontrolResults of Research Activities (RA 4.3) of the EU FP6. These results, complemented by the views of experts in the field of biocontrol consulted at the occasion of meetings of IOBC-wprs, allowed the identification of majors gaps in knowledge and bottlenecks for the successful deployment of biocontrol and lead to the proposition of key issues for future work by the research community, the field of development and prospects for technological improvement by industry.

    The development of new biocontrol products against plant diseases requires screening of high numbers of candidate antagonists. Antagonists for commercial use have to fulfill many different requirements. Besides being active against the specific targeted plant pathogens they must be safe and cost-effective. Important criteria besides pathogenic efficacy are market size, ecological characteristics, production costs, safety, environmental risks, and possibilities for intellectual property protection. A stepwise screening program considering these very different aspects is proposed.     BcatrA was cloned from the plant pathogenic fungus Botryotinia fuckeliana (Botrytis cinerea) and sequenced. Sequence analysis revealed that BcatrA encodes a protein composed of 1562 amino acid residues displaying high similarity with various fungal ATP-binding cassette (ABC) transporters having the (NBF-TM6)2 topology. Expression of BcatrA is barely detectable during normal vegetative growth in liquid substrates. Transcript levels of BcatrA are enhanced in a dose- and time-dependent manner after treatment with cycloheximide or catechol, but not by a number of other drugs or fungicides, including fludioxonil, fenarimol, imazalil, and the plant defense compounds pisatin and resveratrol. Quantitative analysis of BcatrA during the synchronized infection of bean leaves revealed an overaccumulation of the gene transcript at 6, 12 and 24 h post-inoculation, suggesting an involvement of the gene in the first steps of pathogenesis. Functional analysis of BcatrA was performed by targeted gene replacement in a wild-type strain of the fungus, and by overexpression in a mutant of Saccharomyces cerevisiae carrying multiple non-functional multidrug-resistance genes. BcatrA replacement mutants did not show any significant increase in sensitivity to drugs, including inducers of BcatrA transcription, and displayed an unaltered virulence on several common host plants of B. cinerea. However, when expressed in the heterologous system, BcatrA reduced sensitivity to cycloheximide and catechol, thus indicating the ability of the BcatrA product to function as a multidrug transporter.

    Amadori products - Amadoriase I - Browning - Protein glycation

    Amadoriase I is a fructosyl amine oxidase from Aspergillus fumigatus that catalyzes the oxidation of Amadori products (APs) producing glucosone, H 2O2, and the corresponding free amine. All the enzymes of this family discovered so far only deglycate small molecular weight products and are inactive toward large molecular weight substrates, such as glycated BSA or ribonuclease A. Therefore, they cannot be used to reverse protein glycation occurring in diabetes or in foods. In this paper, the effect of Amadoriase I added during the in vitro reaction between glucose and peptides having different polarities or proteins with molecular weights ranging from to 5 to 66 kDa was tested. The formation of APs was monitored by ESI-MS of intact glycated protein or peptides and by measuring the Nε-(1-deoxy-D-fructos-1-yl)-L- lysine and furosine concentrations. Results showed that the formation of APs is reduced up to 80% when peptides and glucose are incubated in the presence of Amadoriase. The effect is more evident for hydrophobic peptides. In protein-glucose systems, the effect was dependent on the molecular weight and steric hindrance being negligible for BSA and at a maximum for insulin, where the formation of APs was reduced up to 60%. These findings indicate new potential applications of Amadoriase I as an efficient tool for inhibiting protein glycation in real food systems.
