    Survival of Ralstonia solanacearum and Ralstonia pseudosolanacearum in drain water

    The survival in drain water of two strains of Ralstonia solanacearum and three strains of Ralstonia pseudosolanacearum, including two strains able to cause wilt in roses, was determined. Water draining from drip-irrigated rock wool mats on which roses were grown was supplemented with the pathogen and survival was monitored at 4, 12, 20 and 28°C for up to 112 days. All strains were able to survive for at least 112 days in drain water at 12 and 20°C, but at 4°C maximum survival was 56 days. At 28°C, the survival period was strain dependent, but was at least 56 days. Populations declined gradually in non-sterile drain water to a low level (maximum 100 cfu mL−1 after 112 days). In sterile drain water (autoclaved prior to addition of populations), no or only a limited decline in populations was found at 112 days, dependent on strain and temperature. Drain water that tested negative for Ralstonia in the dilution plating assay was tested for the presence of cells in a viable but non-culturable state (VBNCs). Tomato plants were inoculated, but no symptoms developed, and plants sampled 22 days post-inoculation were negative in a plating assay. Therefore, no indications were found that VBNCs were present.

    Societies worldwide are investing considerable resources into the safe development and use of nanomaterials. Although each of these protective efforts is crucial for governing the risks of nanomaterials, they are insufficient in isolation. What is missing is a more integrative governance approach that goes beyond legislation. Development of this approach must be evidence based and involve key stakeholders to ensure acceptance by end users. The challenge is to develop a framework that coordinates the variety of actors involved in nanotechnology and civil society to facilitate consideration of the complex issues that occur in this rapidly evolving research and development area. Here, we propose three sets of essential elements required to generate an effective risk governance framework for nanomaterials. (1) Advanced tools to facilitate risk-based decision making, including an assessment of the needs of users regarding risk assessment, mitigation, and transfer. (2) An integrated model of predicted human behavior and decision making concerning nanomaterial risks. (3) Legal and other (nano-specific and general) regulatory requirements to ensure compliance and to stimulate proactive approaches to safety. The implementation of such an approach should facilitate and motivate good practice for the various stakeholders to allow the safe and sustainable future development of nanotechnology.

    The 2011 enterohaemorrhagic Escherichia coli (EHEC) outbreak in Germany casted new light on the potential reservoirs of human pathogenic bacteria (HUPA) other than the ones commonly recognized in animal production chains. Soil, plants and water systems were demonstrated to be environments where HUPA can survive for longer periods in time and where they may acquire genotypic properties from resident microbes, thereby combining traits that allow them to adapt to novel ecosystems. In analogy with zoonosis, we propose the term phytonosis for HUPA strains adapted to the plant environment that are transmitted via consumption to humans to cause disease

    For some microbial species, such as Bacillus anthracis, the etiologic agent of the disease anthrax, correct detection and identification by molecular methods can be problematic. The detection of virulent B. anthracis is challenging due to multiple virulence markers that need to be present in order for B. anthracis to be virulent and its close relationship to Bacillus cereus and other members of the B. cereus group. This is especially the case in environments where build-up of Bacillus spores can occur and several representatives of the B. cereus group may be present, which increases the chance for false-positives. In this study we show the presence of B. anthracis-like bacteria and other members of the B. cereus group in a microbial community within the human environment of the International Space Station and their preliminary identification by using conventional culturing as well as molecular techniques including 16S rDNA sequencing, PCR and real-time PCR. Our study shows that when monitoring the microbial hygiene in a given human environment, health risk assessment is troublesome in the case of virulent B. anthracis, especially if this should be done with rapid, easy to apply and on-site molecular methods.

    In deze bijdrage komen de droge heiden van de Veluwe aan bod. Waarbij aan de hand van twee studiegebieden een nauwgezette analyse van het heidelandschap gepresenteerd wordt. Van de voormalig woeste gronden zijn eigenlijk alleen de allerarmste gronden bewaard gebleven.     Landgoederen spelen vanouds een vooraanstaande rol in het integraal en multifunctioneel inrichten en beheren van de groene ruimte. Zij moeten zich telkens opnieuw vernieuwen om vitaal te blijven. Dat kan alleen als zij zich bewust verhouden tot de Nederlandse verstedelijkte samenleving. Zij moeten ook zorgen dat zij gezien worden om die plek te behouden. Zij zullen zichtbaar moeten maken wat hun rol en waarden zijn voor de maatschappelijke behoeften van hun omgeving. In het project ‘Landgoed in 3D!’ zoeken vooruitstrevende landgoedeigenaren naar een nieuwe visie op de rol van landgoederen.     Epidemiology of dark leaf spot caused by Alternaria brassicicola and Alternaria brassicae in organic seed production of cauliflower

    In organic seed production of Brassica vegetables, infections by Alternaria brassicicola and A. brassicae can cause severe losses of yield and seed quality. Four field experiments with or without artificial inoculation with A. brassicicola were conducted in organically managed seed-production crops of cauliflower cv. Opaal RZ in 2005 and 2006 in the Netherlands. The development of A. brassicicola and A. brassicae on pod tissues and developing seeds was followed and seed quality was assessed. Alternaria brassicicola was externally present on 1·2% of the seeds 14 days after flowering and observed internally within 4 weeks after flowering. In both seasons, seed colonization by the pathogen increased slowly until maturation but sharply increased during maturation. A similar pattern was found for the colonization of pod tissues by A. brassicicola as quantified by TaqMan-PCR. The incidence of A. brassicicola on mature seeds reached 70–90%. Internal colonization was found for 62–80% of the seeds. External and internal seed colonization by A. brassicae was much lower, with incidences below 3%. The quality of harvested seeds was generally low, with less than 80% of seeds able to germinate. Seed quality was not affected by warm water treatments. It was concluded that A. brassicicola and A. brassicae have the potential to infect pods and seeds soon after flowering. For the production of high quality seeds, producers must prevent such early infections. Therefore, new control measures are needed for use in organic cropping systems

    A partial equilibrium trade model with heterogeneous firms is developed and applied to the issue of compliance with the EU food standards in Polish meat production. The model parameters are estimated using a nonlinear least squares method to match the observed patterns of trade. With asymmetrically distributed productivities homogenising standards tend to increase the concentration of production and exports among the more productive and larger firms. Simulation analysis shows the trade and market structure impacts of support programmes in the context of EU accession. The simulations also highlight the importance of the substitution elasticity between varieties and effects of productivity upgrades
