    Multimodal Tracking of Controlled Staphylococcus aureus Infections in Mice

    There is a need to develop diagnostic and analytical tools that allow noninvasive monitoring of bacterial growth and dissemination in vivo. For such cell-tracking studies to hold translational value to controlled human infections, in which volunteers are experimentally colonized, they should not require genetic modification, and they should allow tracking over a number of replication cycles. To gauge if an antimicrobial peptide tracer, 99m Tc-UBI 29-41 -Cy5, which contains both a fluorescent and a radioactive moiety, could be used for such in vivo bacterial tracking, we performed longitudinal imaging of a thigh-muscle infection with 99m Tc-UBI 29-41 -Cy5-labeled Staphylococcus aureus. Mice were imaged using SPECT and fluorescence-imaging modalities at various intervals during a 28 h period. Biodistribution analyses were performed to quantitate radioactivity in the abscess and other tissues. SPECT and fluorescence imaging in mice showed clear retention of the 99m Tc-UBI 29-41 -Cy5-labeled bacteria following inoculation in the thigh muscle. Despite bacterial replication, the signal intensity in the abscess only modestly decreased within a 28 h period: 52% of the total injected radioactivity per gram of tissue (%ID/g) at 4 h postinfection (pi) versus 44%ID/g at 28 h pi (15% decrease). After inoculation, a portion of the bacteria disseminated from the abscess, and S. aureus cultures were obtained from radioactive urine samples. Bacterial staining with 99m Tc-UBI 29-41 -Cy5 allowed noninvasive bacterial-cell tracking during a 28 h period. Given the versatility of the presented bacterial-tracking method, we believe that this concept could pave the way for precise imaging capabilities during controlled-human-infection studies.

    In vivo stability of supramolecular host–guest complexes monitored by dual-isotope multiplexing in a pre-targeting model of experimental liver radioembolization

    Introduction: Cyclodextrin (CD)-based supramolecular interactions have been proposed as nanocarriers for drug delivery. We previously explored the use of these supramolecular interactions to perform targeted hepatic radioembolization. In a two-step procedure the appropriate location of the diagnostic pre-targeting vector can first be confirmed, after which the therapeutic vector will be targeted through multivalent host–guest interactions. Such a procedure would prevent therapeutic errors that come from a mismatch between diagnostic and therapeutic procedures. In the current study we explored the use of dual-isotope imaging to assess the in vivo stability of the formed complex and individual components. Methods: Dual-isotope imaging of the host and guest vectors was performed after labeling of the pre-targeted guest vector, being adamantane (Ad) functionalized macro-aggregated albumin (MAA) particles, with technetium-99 m (99mTc-MAA-Ad). The host vector, Cy50.5CD9PIBMA39, was labeled with indium-111 (111In-Cy50.5CD9PIBMA39). The in situ stability of both the individual vectors and the resulting [MAA-Ad–111In-Cy50.5CD9PIBMA39] complexes was studied over 44 h at 37 °C in a serum protein-containing buffer. In vivo, the host vector 111In-Cy50.5CD9PIBMA39 was administered two hours after local deposition of 99mTc-MAA-Ad in mice. Dual-isotope SPECT imaging and quantitative biodistribution studies were performed between 2 and 44 h post intravenous host vector administration. Results: The individual vectors portrayed <5% dissociation of the radioisotope over the course of 20 h. Dissociation of [MAA-Ad–111In-Cy50.5CD9PIBMA39] complexes remained within a 10–20% range after incubation in serum. In vivo dual-isotope SPECT imaging of host–guest interactions revealed co-localization of the tracer components. Quantitative assessment of the biodistribution revealed that the hepatic accumulation of the host vector nearly doubled between 2 h and 44 h post-injection (from 14.9 ± 6.1%ID/g to 26.2 ± 2.1%ID/g). Conclusions: Assessment of intra-hepatic host–guest complexation was successfully achieved using dual isotope multiplexing, underlining the complex stability that was found in situ (up to 44 h in serum). Overall, the results obtained in this study highlight the potential of supramolecular chemistry as a versatile platform that could advance the field of nanomedicine.

    Roots Partially in Contact with Soil: Analytical Solutions and Approximation in Models of Nutrient and Water Uptake

    Root–soil contact entails a trade-off between uptake opportunities and aeration requirements. A single root in the center of a cylinder of soil has been the standard geometry for which most root-level water and nutrient uptake models have been derived. However, this implies assumptions about complete root–soil contact and regularly spaced, parallel roots that do not conform to the situation in the field. In reality, the frequency distribution of transport distances will differ from what the cylinder model assumes, both by partial root–soil contact and irregular three-dimensional (3-D) distribution. We derived analytical equations describing the transport of water and nutrients to and uptake by roots in partial contact with soil for two extremes: (i) part of each root in contact or (ii) part of all roots in full contact and the rest without. Solutions range from negligible impacts to proportionality of uptake to the part of roots in contact with soil.

    Fluorescent Lectins for Local in Vivo Visualization of Peripheral Nerves

    Damage to peripheral nerves caused during a surgical intervention often results in function loss. Fluorescence imaging has the potential to improve intraoperative identification and preservation of these structures. However, only very few nerve targeting agents are available. This study describes the in vivo nerve staining capabilities of locally administered fluorescent lectin-analogues. To this end WGA, PNA, PHA-L and LEL were functionalized with Cy5 (lambda(ex) (max) 640 nm; lambda(em) (max) 680 nm). Transfer of these imaging agents along the sciatic nerve was evaluated in Thy1-YFP mice (n = 12) after intramuscular injection. Migration from the injection site was assessed in vivo using a laboratory fluorescence scanner and ex vivo via fluorescence confocal microscopy. All four lectins showed retrograde movement and staining of the epineurium with a signal-to-muscle ratio of around two. On average, the longest transfer distance was obtained with WGA-Cy5 (0.95 cm). Since WGA also gave minimal uptake in the lymphatic system, this lectin type revealed the highest potential as a migration imaging agent to visualize nerves.
    Root water uptake as simulated by three water flow models

    The objective of our study was to determine to what extent four root water uptake (RWU) models differed when tested under extreme conditions. Cumulative transpiration patterns were similar, contrary to the spatial extraction patterns. The analysis showed that both soil physical and root physiological factors are important for proper deterministic modeling of RWU. We compared four root water uptake (RWU) models of different complexity that are all embedded in greater soil water flow models. The soil models used were SWAP (one-dimensional), FUSSIM2 (two-dimensional), and RSWMS (three-dimensional). Within SWAP, two RWU functions were utilized (SWAP-macro and SWAP-micro). The complexity of the processes considered in RWU increases from SWAP-macro, to SWAP-micro, to FUSSIM2, to RSWMS. The objective of our study was to determine to what extent the RWU models differed when tested under extreme conditions: low root length density, high transpiration rate, and low water content. Comparison 1 looked at the results of the models for a scenario of transpiration and uptake and Comparison 2 studied compensation mechanisms of water uptake. The uptake scenario pertained to a long dry period with constant transpiration and a single rainfall event. As could be expected, the models yielded different results in Comparison 1, but the differences in cumulative transpiration were modest due to various feedback mechanisms. In Comparison 2, the cumulative effect of different feedback processes were studied. Redistribution of water due to soil pressure head gradients generated by water uptake led to an increase in cumulative transpiration of 32%, and the inclusion of compensation in uptake by the roots resulted in a further increase of 10%. Going from one- to three-dimensional modeling, the horizontal gradients in the soil and root system increased, which reduced the actual transpiration.     In opdracht van Rijkswaterstaat, Directie IJsselmeergebied, is in 1994 een monitoringsprogramma gestart waarin zeldzame vissoorten in het IJsselmeergebied bemonsterd worden. Vanaf 2000 wordt er ook aan de Waddenzeezijde van de Afsluitdijk een monitoringprogramma van diadrome vis uitgevoerd. Het doel van de programma’s is voornamelijk om trends en ontwikkelingen in diadrome vissoorten (fint, harder, houting, grote marene, rivierprik, roofblei, zeeprik, zalm en zeeforel) in het IJsselmeergebied en aan de zoute kant van de Afsluitdijk te beschrijven. Deze monitoring levert inzicht in het voorkomen van diadrome vis en kan gebruikt worden voor de evaluatie van het effect van geplande veranderingen in het spuibeheer op de mogelijkheden voor vistrek en de effectiviteit van een vispassage in het nieuwe spuicomplex in de Afsluitdijk.

    Water uptake by plant roots is an important component of the soil water balance. In this report we studied four water uptake models, of different complexity, that were all embedded in a greater model dealing with transport of water in (an unsaturated) soil. Though also some attention was paid to performance of the routines by themselves, the focus was directed to their functioning as a part of the greater models. We examined the results of two scenarios of potential transpiration and precipitation, comprising a period of 16 days. As could be expected the models yielded different results, but the differences in actual transpiration are modest due to feedback mechanisms.

    In dit rapport worden de gegevens van de passieve monitoring van de visstand in de zoete rijkswateren, die in 2010 zijn verzameld, gepresenteerd. Voor de passieve monitoring wordt op 31 locaties, normaliter in de periode april tot en met november, door beroepsvissers hun vangst op twee manieren geregistreerd. In 2010 zijn binnen de passieve monitoring in totaal 35 inheemse zoetwatervissoorten waargenomen.

    IMARES voert ter ondersteuning van het visserijbeleid en ten behoeve van integraal water- en visstandbeheer een jaarlijkse monitoring uit van de visstand in het IJsselmeer en het Markermeer. De vismonitoring bestaat uit een aantal onderdelen die samen een beeld geven van de visstand in het IJsselmeer en Markermeer. Dit rapport presenteert de gegevens van zowel de actieve monitoring als van de commerciële visserij (aanlandingsgegevens en marktbemonsteringen). Dit rapport geeft een overzicht van alle gegevens die door de jaren heen verzameld zijn. De gegevens dienen als basis voor analyses binnen diverse projecten die leiden tot inzicht over het functioneren van visbestanden en het ecosysteem.

    Sinds 2000 wordt een monitoringprogramma van diadrome vis aan de Waddenzeezijde van de Afsluitdijk uitgevoerd. Het doel van het programma is om trends en ontwikkelingen in de zeldzame diadrome vissoorten (fint, houting, grote marene, rivierprik, zeeprik, zalm en zeeforel) aan de zoute kant van de Afsluitdijk te beschrijven. Deze monitoring levert inzicht in het voorkomen van diadrome vis en kan gebruikt worden voor de evaluatie van het effect van geplande veranderingen in het spuibeheer op de mogelijkheden voor vistrek en de effectiviteit van een vispassage in het nieuwe spuicomplex in de Afsluitdijk.

    Door de sterke afname van de glasaalintrek verkeert de jaarlijkse bemonstering in de problemen. De kosten van het huidige arbeidsintensieve kruisnetprogramma zijn hoog en de sterke afname van de glasaalvangsten brengen de statistische betrouwbaarheid in gevaar. De ontwikkeling van een nieuwe, betrouwbare en betaalbare methode om de jaarlijkse glasaalmonitoring uit te voeren is belangrijk voor het beheer van de in zorgwekkende toestand verkerende aalstand. In 2010 is er aanvullend onderzoek gedaan naar de mogelijkheid lichtvallen in te zetten als methodiek voor het monitoren van absolute dichtheden glasaal. Het onderzoek in 2010 heeft duidelijk aangetoond dat het twijfelachtig is of lichtvallen een goed alternatief zijn voor de huidige kruisnetbemonstering

    Placement of phosphate leads to a more efficient use of a finite resource

    In a desk study and in a climate chamber experiment we explored the rationale behind the low or even negative correlation between recommended P(hosphorus)-application rates and P-off-take in the Netherlands. We hypothesized that especially for short growing vegetable crops it will be necessary to maintain in the early growth stages a high P-concentration in the soil solution, despite the relatively low P-off-take at harvest. Although the daily P-demand of the crop is low immediately after germination, actual P-uptake might be limited by root length. Placement of P is then a strategy to achieve a more efficient P-fertilization. The fact that P-resources in the future will become scarce will encourage such a strategy. The interaction between soil P-fertility, rooting, P-demand and placement of P was explored with a simulation model. Placement turned out to be very effective to realize the required P-uptake in the early growth stages, more effective than increasing the P-soil fertility status. The effect of placement of P-fertilizer was verified in a climate chamber experiment. In a pot experiment P-fertilization by placement (0, 4, 13 kg P ha-1 in 3% of the soil volume) was compared with 87 kg P ha-1 mixed homogeneously through the entire soil volume. Placement stimulated juvenile growth to a great extent in several crops (including carrots, spinach, onion). Placement was equally or even more effective than the broadcasted treatment indicating that P-placement can potentially lead to a large reduction in the use of P-fertilizer. It opens the possibility to maintain crop productivity and quality at lower P-fertility levels of the soil, thereby saving valuable P-resources for the future.

    In dit rapport worden de gegevens van de passieve monitoring van de visstand in de zoete rijkswateren, die in 2009 zijn verzameld, gepresenteerd. Voor de passieve monitoring wordt op 29 locaties, normaliter in de periode april tot en met november, door beroepsvissers hun vangst op twee manieren geregistreerd. Sinds 1993 wordt een vangstregistratie van de commerciële fuikenvisserij op aal bijgehouden. Naast de doelsoort aal worden ook van de bijgevangen vissoorten de aantallen en lengtes geregistreerd.
    From QTL to candidate gene: Genetical genomics of simple and complex traits in potato using a pooling strategy

    Background: Utilization of the natural genetic variation in traditional breeding programs remains a major challenge in crop plants. The identification of candidate genes underlying, or associated with, phenotypic trait QTLs is desired for effective marker assisted breeding. With the advent of high throughput -omics technologies, screening of entire populations for association of gene expression with targeted traits is becoming feasible but remains costly. Here we present the identification of novel candidate genes for different potato tuber quality traits by employing a pooling approach reducing the number of hybridizations needed. Extreme genotypes for a quantitative trait are collected and the RNA from contrasting bulks is then profiled with the aim of finding differentially expressed genes. Results: We have successfully implemented the pooling strategy for potato quality traits and identified candidate genes associated with potato tuber flesh color and tuber cooking type. Elevated expression level of a dominant allele of the ß-carotene hydroxylase (bch) gene was associated with yellow flesh color through mapping of the gene under a major QTL for flesh color on chromosome 3. For a second trait, a candidate gene with homology to a tyrosine-lysine rich protein (TLRP) was identified based on allele specificity of the probe on the microarray. TLRP was mapped on chromosome 9 in close proximity to a QTL for potato cooking type strengthening its significance as a candidate gene. Furthermore, we have performed a profiling experiment targeting a polygenic trait, by pooling individual genotypes based both on phenotypic and marker data, allowing the identification of candidate genes associated with the two different linkage groups. Conclusions: A pooling approach for RNA-profiling with the aim of identifying novel candidate genes associated with tuber quality traits was successfully implemented. The identified candidate genes for tuber flesh color (bch) and cooking type (tlrp) can provide useful markers for breeding schemes in the future. Strengths and limitations of the approach are discussed.

    Uit de open water monitoring blijkt dat de visstand net zoals in voorgaande jaren in het geheel lager is in het Markermeer dan in het IJsselmeer. De vangsten in beide meren worden gedomineerd door pos, aal, snoekbaars, baars, spiering, blankvoorn, brasem en bot. Het verloop van deze bemonsterde bestanden wordt uitvoerig in het rapport besproken met behulp van de resultaten uit de open water monitoring en de aanlandingsgegevens en marktbemonsteringen. Daarnaast worden de vangsten van Flora- en Faunawet soorten en limnofiele soorten binnen de actieve monitoring besproken
