    CRISPR/Cas9-targeted mutagenesis of the tomato susceptibility gene PMR4 for resistance against powdery mildew

    CRISPR/Cas9 - PMR4 - Powdery mildew - Susceptibility gene - Targeted mutagenesis

    Background: The development of CRISPR/Cas9 technology has facilitated targeted mutagenesis in an efficient and precise way. Previously, RNAi silencing of the susceptibility (S) gene P owdery M ildew R esistance 4 (PMR4) in tomato has been shown to enhance resistance against the powdery mildew pathogen Oidium neolycopersici (On). Results: To study whether full knock-out of the tomato PMR4 gene would result in a higher level of resistance than in the RNAi-silenced transgenic plants we generated tomato PMR4 CRISPR mutants. We used a CRISPR/Cas9 construct containing four single-guide RNAs (sgRNAs) targeting the tomato PMR4 gene to increase the possibility of large deletions in the mutants. After PCR-based selection and sequencing of transformants, we identified five different mutation events, including deletions from 4 to 900-bp, a 1-bp insertion and a 892-bp inversion. These mutants all showed reduced susceptibility to On based on visual scoring of disease symptoms and quantification of relative fungal biomass. Histological observations revealed a significantly higher occurrence of hypersensitive response-like cell death at sites of fungal infection in the pmr4 mutants compared to wild-type plants. Both haustorial formation and hyphal growth were diminished but not completely inhibited in the mutants. Conclusion: CRISPR/Cas-9 targeted mutagenesis of the tomato PMR4 gene resulted in mutants with reduced but not complete loss of susceptibility to the PM pathogen On. Our study demonstrates the efficiency and versatility of the CRISPR/Cas9 system as a powerful tool to study and characterize S-genes by generating different types of mutations.

    The identification of immune receptors in crop plants is time-consuming but important for disease control. Previously, resistance gene enrichment sequencing (RenSeq) was developed to accelerate mapping of nucleotide-binding domain and leucine-rich repeat containing (NLR) genes. However, resistances mediated by pattern recognition receptors (PRRs) remain less utilized. Here, our pipeline shows accelerated mapping of PRRs. Effectoromics leads to precise identification of plants with target PRRs, and subsequent RLP/K enrichment sequencing (RLP/KSeq) leads to detection of informative single nucleotide polymorphisms that are linked to the trait. Using Phytophthora infestans as a model, we identified Solanum microdontum plants that recognize the apoplastic effectors INF1 or SCR74. RLP/KSeq in a segregating Solanum population confirmed the localization of the INF1 receptor on chromosome 12, and led to the rapid mapping of the response to SCR74 to chromosome 9. By using markers obtained from RLP/KSeq in conjunction with additional markers, we fine-mapped the SCR74 receptor to a 43-kbp G-LecRK locus. Our findings show that RLP/KSeq enables rapid mapping of PRRs and is especially beneficial for crop plants with large and complex genomes. This work will enable the elucidation and characterization of the nonNLR plant immune receptors and ultimately facilitate informed resistance breeding.

    Effective elements of care-physical activity initiatives for adults with a low socioeconomic status: A concept mapping study with health promotion experts

    In care-physical activity (care-PA) initiatives, primary care and sports are combined to stimulate PA among adults at risk of lifestyle related diseases. Preliminary results from Dutch care-PA initiatives for adults with a low socioeconomic status (SES) indicate a decrease in participants’ body weight and an improved quality of life, however, the elements that make these initiatives successful are yet to be identified. In total, 19 Dutch health promotion experts participated in our concept mapping (CM) of the effective elements of care-PA initiatives for adults with a low SES. The experts identified 111 effective elements of these initiatives, which were grouped into 11 clusters, focusing on: 1) approaching participants within the care-PA initiative, 2) barriers experienced throughout the initiative, 3) long-term implementation, 4) customizing the care-PA initiative to the target population, 5) social support, 6) structure and guidance, 7) the professionals within the care-PA initiative, 8) the accessibility of the care-PA initiative, 9) targeted behaviour and progression, 10) recruitment and administration, and 11) intersectoral collaboration. CM was useful for creating a valuable overview of these effective elements. Our results could be used to improve the development and implementation of future care-PA initiatives for adults with a low SES.

    A review on the genetic resources, domestication and breeding history of spinach (Spinacia oleracea L.)

    This paper addresses the genetic resources, domestication and breeding history of spinach as a comprehensive review of these crop aspects is currently unavailable. It is shown that the availability of genetic resources of wild relatives belonging to the primary gene pool is currently very limited, which hampers breeding and research activities. Therefore, new collecting expeditions are clearly warranted. The domestication of spinach is discussed on the basis of its presumed migration routes and the traits that were probably involved in the domestication syndrome. Spinach is thought to have domesticated in former Persia. Migration then occurred eastwards to China and westwards to Europe, but additional genetic data are needed to reveal the most likely migration routes. Morphological changes in pistillate flowers and loss of dormancy are identified as the main traits involved in the domestication syndrome of spinach. To a large extent we could re-construct the relationships between spinach cultivars that were developed until the 1950s, but this appeared difficult for the more recent cultivars due to intellectual property protection by breeding companies. Resistance against downy mildew has been the main breeding target in spinach. The introgression of NBS-LRR resistance genes from wild relatives is the major strategy to develop downy mildew resistant cultivars. However, the use of loss-of-function alleles of susceptibility genes may provide a more durable strategy to develop resistant cultivars. So far, abiotic resistance and quality traits have received minor attention in spinach research and breeding. This is expected to change considering the potential effects of climate change on these traits.

    Tomato yellow leaf curl virus (TYLCV), a begomovirus, causes large yield losses and breeding for resistance is an effective way to combat this viral disease. The resistance gene Ty-1 codes for an RNA-dependent RNA polymerase and has recently been shown to enhance transcriptional gene silencing of TYLCV. Whereas Ty-1 was earlier shown to also confer resistance to a bipartite begomovirus, here it is shown that Ty-1 is probably generic to all geminiviruses. A tomato Ty-1 introgression line, but also stable transformants of susceptible tomato cv. Moneymaker and Nicotiana benthamiana (N. benthamiana) expressing the Ty-1 gene, exhibited resistance to begomoviruses as well as to the distinct, leafhopper-transmitted beet curly top virus, a curtovirus. Stable Ty-1 transformants of N. benthamiana and tomato showed fewer symptoms and reduced viral titres on infection compared to wild-type plants. TYLCV infections in wild-type N. benthamiana plants in the additional presence of a betasatellite led to increased symptom severity and a consistent, slightly lowered virus titre relative to the high averaged levels seen in the absence of the betasatellite. On the contrary, in Ty-1 transformed N. benthamiana viral titres increased in the presence of the betasatellite. The same was observed when these Ty-1-encoding plants were challenged with TYLCV and a potato virus X construct expressing the RNA interference suppressor protein βC1 encoded by the betasatellite. The resistance spectrum of Ty-1 and the durability of the resistance are discussed in light of antiviral RNA interference and viral counter defence strategies.

    Background & aims: Malnutrition in older adults results in significant personal, social, and economic burden. To combat this complex, multifactorial issue, evidence-based knowledge is needed on the modifiable determinants of malnutrition. Systematic reviews of prospective studies are lacking in this area; therefore, the aim of this systematic review was to investigate the modifiable determinants of malnutrition in older adults. Methods: A systematic approach was taken to conduct this review. Eight databases were searched. Prospective cohort studies with participants of a mean age of 65 years or over were included. Studies were required to measure at least one determinant at baseline and malnutrition as outcome at follow-up. Study quality was assessed using a modified version of the Quality in Prognosis Studies (QUIPS) tool. Pooling of data in a meta-analysis was not possible therefore the findings of each study were synthesized narratively. A descriptive synthesis of studies was used to present results due the heterogeneity of population source and setting, definitions of determinants and outcomes. Consistency of findings was assessed using the schema: strong evidence, moderate evidence, low evidence, and conflicting evidence. Results: Twenty-three studies were included in the final review. Thirty potentially modifiable determinants across seven domains (oral, psychosocial, medication and care, health, physical function, lifestyle, eating) were included. The majority of studies had a high risk of bias and were of a low quality. There is moderate evidence that hospitalisation, eating dependency, poor self-perceived health, poor physical function and poor appetite are determinants of malnutrition. Moderate evidence suggests that chewing difficulties, mouth pain, gum issues co-morbidity, visual and hearing impairments, smoking status, alcohol consumption and physical activity levels, complaints about taste of food and specific nutrient intake are not determinants of malnutrition. There is low evidence that loss of interest in life, access to meals and wheels, and modified texture diets are determinants of malnutrition. Furthermore, there is low evidence that psychological distress, anxiety, loneliness, access to transport and wellbeing, hunger and thirst are not determinants of malnutrition. There appears to be conflicting evidence that dental status, swallowing, cognitive function, depression, residential status, medication intake and/or polypharmacy, constipation, periodontal disease are determinants of malnutrition. Conclusion: There are multiple potentially modifiable determinants of malnutrition however strong robust evidence is lacking for the majority of determinants. Better prospective cohort studies are required. With an increasingly ageing population, targeting modifiable factors will be crucial to the effective treatment and prevention of malnutrition.

    Background
    In care-physical activity (care-PA) initiatives, primary care and sports collaborate to stimulate PA among adults at risk of lifestyle related diseases. Preliminary results of Dutch care-PA initiatives for low socioeconomic status (SES) adults indicate that these initiatives successfully lower participants’ body weight and improve quality of life. However, insight into elements that make these initiatives work is lacking. Therefore, this research aims to unravel the effective elements of care-PA initiatives for low SES adults.

    Methods
    Concept Mapping (CM) was used as tool to identify and cluster the effective elements. Nineteen Dutch health promotion experts individually listed as many elements as they felt were of importance to the effectiveness of care-PA initiatives. Next, each expert was asked to cluster the elements and to score them on importance. Then, CS Global MAX software was used for multidimensional scaling and a hierarchical cluster analysis to develop a cluster map. Finally, the cluster map was presented, discussed, and refined in a group meeting with 11 of the experts.

    Results
    The experts came up with 113 unique effective elements of care-PA initiatives for low SES adults, clustered into 11 clusters: 1) approach of professionals, 2) barriers experienced during the programme, 3) local embedding, 4) customisation of the programme to target population, 5) social support, 6) methods within the programme, 7) competencies of professionals, 8) accessibility of the programme, 9) actions within the programme, 10) recruitment of participants, and 11) intersectoral collaboration.

    Conclusions
    A valuable overview of the effective elements of care-PA initiatives for low SES adults was created. The results can be used to improve existing care-PA initiatives and to develop new ones targeted at low SES adults at risk of lifestyle related diseases. This may eventually help to reduce health inequalities between low and high SES adults.

    Key messages
    Concept mapping has been a useful group-based tool to obtain information on the effective elements of care-PA initiatives, in which individual input from health promotion experts has been collected.
    The overview of effective elements of care-PA initiatives for low SES adults as presented in this study is valuable for the development of care-PA initiatives specifically targeted at low SES adults.

    Soil organisms, including earthworms, are a key component of terrestrial ecosystems. However, little is known about their diversity, their distribution, and the threats affecting them. We compiled a global dataset of sampled earthworm communities from 6928 sites in 57 countries as a basis for predicting patterns in earthworm diversity, abundance, and biomass. We found that local species richness and abundance typically peaked at higher latitudes, displaying patterns opposite to those observed in aboveground organisms. However, high species dissimilarity across tropical locations may cause diversity across the entirety of the tropics to be higher than elsewhere. Climate variables were found to be more important in shaping earthworm communities than soil properties or habitat cover. These findings suggest that climate change may have serious implications for earthworm communities and for the functions they provide.

    Die Landwirtschaft in Deutschland, Europa und weltweit befindet sich in einem dramatischen Umbruchprozess. Einerseits bieten Liberalisierung und Marktöffnung, neue technologische Entwicklungen, die wachsende Weltbevölkerung und neue Kundenwünsche vielen landwirtschaftlichen Betrieben neue Möglichkeiten. Andererseits stehen viele Landwirtinnen und Landwirte in internationaler Konkurrenz, sind zum betrieblichen Wachstum mit hohem Investitionsrisiko gezwungen, sind Teil strikt regulierter Wertschöpfungsketten und müssen steigenden gesellschaftlichen Ansprüchen genügen. Zugleich machen sich viele Menschen Sorgen, dass die Prozesse der Rationalisierung und Modernisierung der landwirtschaftlichen Produktion auf Kosten von Natur-, Umwelt- und Tierschutz gehen. Auf vielen Ebenen wird daher bereits nach einer neuen Verständigung darüber gesucht, was die Gesellschaft von der Landwirtschaft erwartet, und welche Unterstützung die Landwirtinnen und Landwirte im Gegenzug dafür erwarten dürfen. Vor diesem Hintergrund entwickelt das vorliegende Open Access-Buch eine wissenschaftlich fundierte Kritik der Umweltauswirkungen der Landwirtschaft und der politischen Logik der Agrarpolitik. Es präsentiert ein neues Leitbild mit konkreten Indikatoren sowie alternative strategische Handlungsoptionen. Die umfassende Analyse wird zu einem Vorschlag für eine neue Architektur der europäischen Agrarpolitik gebündelt, die eine breite Diskussion um einen Neuen Gesellschaftsvertrag für die Landwirtschaft in Deutschland und Europa anstoßen soll.

    From being a smallholder-based, food-producing country covering its basic needs, Iraq and the Kurdistan Region in Iraq (KRI) have become major importers of food. The sustainability of the agricultural sector has been systematically undermined by conflict, neglect, and mismanagement, as a result of which the capacity of its farmers to feed the population declined. Even though local policymakers, the international community, and the international organisations emphasise the potential of agriculture for food production, job creation, and income generation, they also tend to consider the current food system problematic because of an alleged low productivity that they relate to the existing smallholder system. For them, such system poses a lack of competences and skills of farmers, and a subsistence production orientation. This approach culminated in a policy-making process that offered land and water for capital investments, and thus neglecting the potentials and competencies of (small-scale) farmers. The concomitant neglect of the human dimension of agriculture, namely the family farm, is essentially the continuation of an economically and ecologically high-risk approach that may lead to a further decline of the sector’s ability to produce food for the local market.
    Effective elements of care-physical activity initiatives for citizens with a low socioeconomic status

    PURPOSE Care-physical activity (care-PA) initiatives involve collaborations between the primary care and the sports sector, with the aim to stimulate PA among citizens at risk for lifestyle related diseases. Preliminary results of Dutch care-PA initiatives specifically designed for adults with a low socioeconomic status (SES) indicate that these initiatives are successful in lowering body weight and enhancing quality of life among participants. While this is encouraging, insights into the elements that make these initiatives effective would be useful for the development and implementation of new initiatives. Therefore, this research aims to unravel the effective elements of care-PA initiatives for low SES adults.
    METHODS Concept Mapping (CM) was used as a tool to identify and cluster the effective elements. Nineteen Dutch health promotion experts participated in the CM procedure. First, each expert individually listed as many elements as they thought were of critical importance to the effectiveness of care-PA initiatives. Next, the researchers created a list of all elements and asked each expert to cluster the elements, and to score them on importance. Then, CS Global MAX software was used to perform multidimensional scaling and a hierarchical cluster analysis to develop a cluster map. Finally, during a group meeting with 11 of the experts, the cluster map was presented, discussed, and refined.
    RESULTS The experts came up with 125 unique effective elements of care-PA initiatives for low SES citizens. After multiple adaptations, e.g. merging and splitting of elements and clusters, as result of input from the respondents during the group meeting, 113 unique effective elements were clustered into 11 clusters: 1) intersectoral collaboration, 2) local embedding, 3) barriers experienced during the programme, 4) customisation of the programme to the target population, 5) accessibility of the programme, 6) recruitment of participants, 7) competencies of the professionals, 8) approach of the professional, 9) methods within the programme, 10) actions within the programme, and 11) social support.
    CONCLUSIONS The findings provide a valuable overview of the effective elements of care-PA initiatives for low SES citizens. The results can be used to develop and implement additional care-PA initiatives targeted at low SES citizens, and may eventually help to reduce health inequalities between low and high SES citizens.
